Sleep deprivation%Propofol%Ciwujia injection
目的 探讨丙泊酚联合刺五加注射液对睡眠剥夺模型大鼠的睡眠影响及机制.方法 睡眠剥夺模型SD大鼠随机分组为四组:空白对照组、丙泊酚组、刺五加组、丙泊酚-刺五加组.采用中缝背核(DRN)核团微量注射、多道睡眠描记、免疫组织化学技术和胆碱能受体放射性配基分析技术,观察大鼠在使用丙泊酚及刺五加注射液后睡眠生理行为、中缝核5-羟色胺(5-HT)能阳性神经元细胞数目,以及皮质神经细胞的胆碱能受体位点数变化.结果 丙泊酚组、刺五加组、丙泊酚-刺五加组的慢波睡眠(SWS)各期睡眠时间显著延长,联合应用后延长效果更显著,而觉醒期(w期)时间明显缩短(P<0.05);各组与对照组比较,大鼠大脑皮层5-HT阳性染色细胞明显增多,平均积分光密度值升高(P<0.05),联合应用后效果更显著.且多重比较显示,丙泊酚组和刺五加组问差异无显著性(P>0.05),其余各组间差异均存在显著性(P值均<0.05).各组间大鼠海马和皮层胆碱能受体位点数比较差异无统计学意义(P值分别为0.081、0.065、0.071).结论 应朋丙泊酚、刺五加、丙泊酚联合刺五加背侧中缝核团微量注射可改善睡眠剥夺大鼠的睡眠.具体机制可能为通过增加大脑皮质细胞内5-HT能阳性物质发挥作用.海马和皮层胆碱能受体位点数改变可能不参与其作用机制.

목적 탐토병박분연합자오가주사액대수면박탈모형대서적수면영향급궤제.방법 수면박탈모형SD대서수궤분조위사조:공백대조조、병박분조、자오가조、병박분-자오가조.채용중봉배핵(DRN)핵단미량주사、다도수면묘기、면역조직화학기술화담감능수체방사성배기분석기술,관찰대서재사용병박분급자오가주사액후수면생리행위、중봉핵5-간색알(5-HT)능양성신경원세포수목,이급피질신경세포적담감능수체위점수변화.결과 병박분조、자오가조、병박분-자오가조적만파수면(SWS)각기수면시간현저연장,연합응용후연장효과경현저,이각성기(w기)시간명현축단(P<0.05);각조여대조조비교,대서대뇌피층5-HT양성염색세포명현증다,평균적분광밀도치승고(P<0.05),연합응용후효과경현저.차다중비교현시,병박분조화자오가조문차이무현저성(P>0.05),기여각조간차이균존재현저성(P치균<0.05).각조간대서해마화피층담감능수체위점수비교차이무통계학의의(P치분별위0.081、0.065、0.071).결론 응붕병박분、자오가、병박분연합자오가배측중봉핵단미량주사가개선수면박탈대서적수면.구체궤제가능위통과증가대뇌피질세포내5-HT능양성물질발휘작용.해마화피층담감능수체위점수개변가능불삼여기작용궤제.
Objective To explore the effects of propofol combined with ciwujia injection on rats with sleep deprivation.Methods SD rats with sleep deprivation were randomly divided into placebo group,propofol group,ciwujia group,and propofol/ciwujia group.Microinjection into DRN,polysomnography (PSG),immunohistochemistrary,and the binding site (Bmax) and the affinity (KD) in the chonlinergic receptors were used in the experiment.The changes of sleep-awaking cycle,5-HT positive neurons,and cholinergic receptors of the cortical neural cells were observed in all the rats.Results The sleeping time of SWS elongated and phase W shortened noticeably in propofol group,ciwujia group,and propofol/ciwujia group (P<0.05).The number of 5-HT positive neurons increased,and no significant change in the number of cholinergic receptors of the,cortical neural cells were found in the latter three groups (P= 0.081,0.065,and 0.071; respectively) Conclusions Combination therapy with propofol and ciwujia and propofol can improve sleep in rats with sleep deprivation,whose mechanism may be the increase in 5-HT positive neurons of the cortical neural cells.
