非小细胞肺癌(non-small cell lung cancer,NSCLC)是严重危害人类健康的常见恶性肿瘤.表皮生长因子受体(epidermal growth factor receptor,EGFR)属于酪氨酸激酶受体,在NSCLC中发现EGFR酪氨酸激酶区常发生各种突变,这些突变和酪氨酸激酶抑制剂(TKI)的疗效密切相关.因此,EGFR突变成为NSCLC患者应用TKI药物强预测因子,临床筛选EGFR突变人群行TKI药物靶向治疗具有重要的临床指导意义.目前用于EGFR突变检测的方法有:直接测序法、RT-PCR、ARMS、DHPLC、HRM、突变富集PCR、PCR-SSCP、PCR-RFLP、微数字PCR、PCR-LDR、荧光偏振法、毛细管电泳法、PNA-LNA等.

Non-small cell lung cancer (NSCLC) is the most common cancer severely endangering human health.Epidermal growth factor receptor (EGFR) is a tyrosine kinase receptor.Lots of mutations in the area of tyrosine kinase have been found in NSCLC,and the mutations are highly correlated with the clinical response of tyrosine kinase inhibitor (TKI).Therefore,EGFR mutations have been the strong predictor for TKI.Screening people of EGFR mutations for targeted therapy has important clinical significance.At present,the detection technology of EGFR mutations include sequencing,RT-PCR,ARMS,DHPLC,HRM,mutant-enriched PCR,PCR-SSCP,PCR-RFLP,microfluidics digital PCR,PCR-LDR,FP,CE,PNA-LNA,and so on.
