目的 观察精子蛋白(SP)32/OY-TES-1在视网膜母细胞瘤(RB)组织中的表达.方法 病理科收检并经病理学检查确诊的30例RB患儿的石蜡标本纳入研究.采用逆转录-聚合酶链反应(RT-PCR)技术检测15例RB组织、12例非肿瘤病变视网膜组织及22例眼部正常组织中SP32/OY-TES-1mRNA的表达;免疫组织化学技术对30例RB组织和24例正常视网膜组织中SP32/OY-TES-1蛋白的表达进行检测,对比分析SP32/OY-TES-1 mRNA和蛋白表达与RB患儿年龄、性别、肿瘤大小、肿瘤分化程度和临床分期的相互关系.结果 RB组织、非肿瘤病变视网膜组织、眼部正常组织SP32/OY-TES-1mRNA的表达率分别为86.7%、16.7%和36.4%.不同性别(χ2=0.744)、年龄≤2岁和>2岁(χ2=1.178)、临床分期(χ2=1.188)、肿瘤大小(χ2=0.216)、肿瘤分型(χ2=1.885)之间SP32/OY-TES-1基因阳性表达率比较,差异无统计学意义(P>0.05).SP32/OY-TES-1蛋白在RB中的表达率为73.3%,在正常视网膜组织中则不表达.不同性别(χ2=1.000)、年龄≤2岁和>2岁(χ2=0.403)、不同肿瘤大小(χ2=2.274)、不同肿瘤分型(χ2=0.138)之间SP32/OY-TES-1蛋白阳性表达率比较,差异无统计学意义(P>0.05);不同临床分期(χ2=6.193)、视神经是否浸润(χ2=4.535)之间SP32/OY-TES-1蛋白阳性表达率比较,差异有统计学意义(P<0.05).结论 SP32/OY-TES-1高表达于RB组织中;SP32/OY-TES-1蛋白在RB组织中的表达率与视神经浸润和临床分期存在密切的关系.

Objective To observe the expression of sperm protein (SP) 32/OY-TES-1 in retinoblastoma (RB). Methods Thirty paraffin specimens of pathologically confirmed RB eyeballs were investigated in this study. The SP32/OY-TES-1 mRNA expression of 15 RB tissues, 12 non-tumor retinal tissues and 22 normal eye tissues were detected by reverse transcription polymerase chain reaction (RT-PCR). The SP32/OY-TES-1 protein expression of 30 RB tissues and 24 normal retinal tissues were examined by immunohistochemistry. The relationship between SP32/OY-TES-1 mRNA, protein expression and age, gender, tumor size, tumor differentiation, clinical stage of RB children were analyzed. Results The expression rate of SP32/OY-TES-l mRNA in RB tissues, non-tumor retinal tissues and normal eye tissues were 86.7%, 16. 7% and 36. 4% respectively. Compared the expression rate of SP32/OY-TES-1mRNA in different gender (χ2 = 0.744 ), age (χ2 = 1.178 ), clinical stage (χ2 = 1. 188 ), tumor size (χ2 =0. 216) ,tumor differentiation (χ2 = 1. 885),the differences were not statistically significant (P>0. 05). The expression rate of SP32/OY-TES-1 protein in RB tissues was 73.3% and no expression in normal retinal tissues. Compared the expression rate of SP32/OY-TES-1 protein in different gender (χ2= 1.000), age with ≤ two years and > two years (χ2 = 0. 403), tumor size (χ2 = 2. 274), tumor differentiation (χ2=0. 138), the differences were not statistically significant (P>0.05) ;but in clinical stage (χ2 =6. 193) ,with or without optic nerve infiltration (χ2= 4. 535), the differences were statistically significant (P<0.05).Conclusions SP32/OY-TES-1 is highly expressed in RB. The expression rate of SP32/OY-TES-1 is related to optic nerve infiltration and clinical stage of RB.
