目的:研究高功率微波 (high power microwave,HPM)对心肌细胞和心脏间质细胞生存能力和细胞活性的影响. 方法:实验选用出生后 1~ 3 d的二级 Wistar大鼠,体外培养 Wistar大鼠乳鼠心肌细胞和心脏间质细胞,于 HPM辐照后,用流式细胞仪检测细胞凋亡和坏死;用 MTT实验测定细胞活力. 结果: HPM明显影响心肌细胞和间质细胞功能和形态结构,辐照后细胞活力降低,凋亡和坏死.辐照后 6 h,心肌细胞和间质细胞凋亡率分别为( 36.76± 5.12)%和( 42.38± 6.0)%,坏死率分别为( 34.14± 6.5)%和( 22.98± 7.4)%,与对照组相比,有显著的统计学意义( P< 0.001). 结论:心脏是微波辐照的靶器官之一, HPM可对心肌细胞和间质细胞功能和形态产生明显影响.结果提示在探讨 HPM对心脏损伤时,应同时关注对心脏间质细胞的影响.

AIM:To study the effect of high power microwave(HPM) on the viability and activity of cardiac myocytes and cardiac interstitial cells. METHODS:Cardiac myocytes and cardiac interstitial cells of Wistar rats(1- 3 d Wistar rats) were cultured in vitro.Apoptosis and necrosis of cells were detected by flow cytometer and the activity of cells was assayed by MTT after cardiac myocytes and cardiac interstitial cells irradiated by HPM. RESULTS:HPM could influence the function and morphology of the two kinds of cells obviously.The activity of cells decreased after irradiation and apoptosis and necrosis of cells occurred.After irradiation 6 h,the apoptosis rate of cardiac myocytes and cardiac interstitial cells were( 36.76± 5.12)% and( 42.38± 6.0)% respectively;the necrosis rate of cardiac myocytes and cardiac interstitial cells were( 34.14± 6.5)% and( 22.98± 7.4)% respectively.Compared with the control group,there was obviously statistical significance( P< 0.001) . CONCLUSION:Heart is one of target organs to microwave irradiation.HPM can influence the function and morphology of cardiac myocytes and cardiac interstitial cells obviously.The results suggested that we should pay more attention to the effect of HPM on cardiac interstitial cells when the effect of HPM on injury of heart was studied.
