目的 观察电针对海洛因依赖大鼠戒断症状及中央灰质背侧部(CGd)、被盖背外侧核(ldt)和蓝斑(LC)等脑区一氧化氮合酶(Nitricoxide synthase ,NOS)阳性神经元表达的影响.方法 SD大鼠随机分为空白对照组、戒断组、捆绑组和电针组.大鼠逐日递增皮下注射海洛因9d建立海洛因依赖模型,同时每天进行电针干预.纳洛酮激发戒断症状、评分.还原型辅酶Ⅱ黄递酶组织化学法染色,测定相关脑区NOS阳性神经元表达.结果 空白对照组纳洛酮激发戒断症状总评分为(5.4±1.3)分,戒断组为(18.7±3.9)分,2组间差异有显著性(P<0.05),电针组为(10.7±3.6)分,和戒断组相比差异有显著性(P<0.05),捆绑组为(14.3±4.8)分,与戒断组比较差异无显著性(P>0.05),表明电针可以有效减轻海洛因依赖大鼠纳洛酮激发戒断症状;空白对照组CGd、ldt和LC 等脑区NOS阳性神经元表达水平均较低,戒断组上述部位NOS阳性神经元表达较空白对照组均显著增多(P<0.05),而电针处理明显减少戒断大鼠这些部位NOS阳性神经元的表达(P<0.05).结论 电针能有效控制海洛因依赖大鼠戒断症状,其作用机制可能与其调节相关脑区NOS阳性神经元表达有关.

목적 관찰전침대해락인의뢰대서계단증상급중앙회질배측부(CGd)、피개배외측핵(ldt)화람반(LC)등뇌구일양화담합매(Nitricoxide synthase ,NOS)양성신경원표체적영향.방법 SD대서수궤분위공백대조조、계단조、곤방조화전침조.대서축일체증피하주사해락인9d건립해락인의뢰모형,동시매천진행전침간예.납락동격발계단증상、평분.환원형보매Ⅱ황체매조직화학법염색,측정상관뇌구NOS양성신경원표체.결과 공백대조조납락동격발계단증상총평분위(5.4±1.3)분,계단조위(18.7±3.9)분,2조간차이유현저성(P<0.05),전침조위(10.7±3.6)분,화계단조상비차이유현저성(P<0.05),곤방조위(14.3±4.8)분,여계단조비교차이무현저성(P>0.05),표명전침가이유효감경해락인의뢰대서납락동격발계단증상;공백대조조CGd、ldt화LC 등뇌구NOS양성신경원표체수평균교저,계단조상술부위NOS양성신경원표체교공백대조조균현저증다(P<0.05),이전침처리명현감소계단대서저사부위NOS양성신경원적표체(P<0.05).결론 전침능유효공제해락인의뢰대서계단증상,기작용궤제가능여기조절상관뇌구NOS양성신경원표체유관.
Objective To investigate the effects of electroacupuncture(EA) on the withdrawal symptoms of heroin dependent rats and the nitricoxide synthase(NOS) expression in encephalic regions including central gray dorsal part( CGd),laterodorsal tegmental neucleus(ldt) and locus ceruleus(LC).Methods SD rats were randomly divided into normal control,wihdrawal,restrainted and EA group.Rats were injected subcutaneously with heroin according a gradually increasing dose scheme for 9 days to establish model of heroin dependence;rats were treated with EA during the heroin treatment.Naloxone(4 mg/kg) was injected intraperitoneally four hours after the last heroin injection.Total withdrawal symptoms scores were assessed within half an hour just after naloxone injection.Nicotinamide adenosine dinucleotide phosphate diaphorase (NADPH-d) staining was used to determine the expression of positive neurons of nitric oxide synthase(NOS) in the related encephalic regions.Results The total scores of withdrawal symptoms precipitated by naloxone in normal control group was (5.4±1.3)points and that of withdrawal group was (18.7±3.9)points.There was significant difference between the two groups(P<0.05).The EA group's was (10.7±3.6) points.There was significant difference between the EA and withdrawal group(P<0.05).The expression of Nitricoxide synthase positive neuron in CGd,ldt and LC in normal control group was rather low,but that of withdrawal group was significant increased(P<0.05).There was dramatical decrease in EA group contrast to the withdrawal group(P<0.05).There was no difference between the restrainted group and the withdrawal group(P>0.05).Conclusion EA could inhibit the withdrawal symptoms of heroin dependence rats,whose mechanism might be involved in the alteration of expression of NOS positive neuron in related encephalic regions.
