目的 观察SD大鼠帕金森病(PD)模型胃肠功能障碍及肌间神经丛多巴胺能、胆碱能神经元的变化.方法 6-羟基多巴胺(6-OHDA)诱导制备PD大鼠模型.测定大鼠1 h粪便排出量、粪便含水量和餐后2 h胃固体食物残留率;记录离体胃肠平滑肌收缩活动;免疫组织化学法检测胃肠神经丛酪氨酸羟化酶(TH)、乙酰胆碱转移酶(ChAT)表达;逆转录-聚合酶链反应(RT-PCR)检测胃肠组织TH、ChAT mRNA水平.结果 6-OHDA组1h粪便湿重、于重、粪便含水量分别为(1.047±0.124)g、(0.812±0.093)g、(22.442±1.180)%,较对照组减少(P<0.01);胃食物残留率为(72.316±3.834)%.较对照组增加(P<0.01);胃、肠平滑肌肌条收缩幅度分别是(0.824 ±0.152)g、(1.541±0.103)g,较对照组降低(P<0.01);胃、肠肌间神经丛TH阳性区平均积分吸光度分别是147.74±13.92、150.45±10.09,显著高于对照组(P<0.01);ChAT阳性区平均积分吸光度分别是109.36±3.37、104.73±5.93,与对照组比较差异无统计学意义(P>0.05);胃、肠组织TH mRNA相对表达量分别是0.662±0.011、0.585±0.012,显著低于对照组(P<0.01);ChAT mRNA相对表达量分别是1.157 ±0.037、1.154±0.031,与对照组比较差异无统计学意义(P>0.05).结论 PD大鼠胃排空延迟和便秘可能与肠神经系统多巴胺含量增多、胆碱能神经递质相对不足有关.

Objective To observe the gastrointestinal dysfunction and the changes of dopaminergic and cholinergic neurons in the myenteric plexuses of Sprague-Dawley (SD) rats model with parkinson' s disease (PD). Methods A rat model of PD was induced by 6-hydroxydopamine (6-OHDA). The one-h fecal output, stool water content and residual solid food in the stomach at 2 h after feeding were measured.Contractile activity of isolated antrum and colon smooth muscle were recorded. Changes in tyrosine hydroxylase (TH) and choline acetyltransferase (ChAT) in the gastrointestinal myenteric plexus were studied by immunohistochemistry. Changes in TH and ChAT mRNA levels in the gastric antrum and colon tissue were determined by reverse transcription-polymerase chain reaction ( RT-PCR). Results In the 6-OHDA group, the wet weight, dry weight and water content of the feces within 1 h were (1. 047 ± 0. 124) g,(0. 812 ±0.093) g, (22.442 ±1. 180)% respectively, which were lower than in the control group (P<0.01); The percentage of residual solid food was (72. 316 ±3. 834)% , that was higher than in the control group (P <0. 01); The contractile amplitude of gastric antrum and colon smooth muscle strips was (0. 824 ±0. 152) g and (1. 541 ±0. 103) g respectively, that were lower than in the control group (P <0.01);The average integrated absorbance values of TH-positive areas in the myenteric plexus of gastric antrum and colon tissue were 147. 74 ± 13. 92 and 150. 45 ± 10. 09 respectively, that were higher than in the control group (P <0. 01); There was no significant difference in ChAT immunoreactivity between 6-OHDA group (109. 36 ± 3. 37 and 104. 73 ± 5. 93) and control group ( P > 0. 05); The mRNA expression levels of TH in the gastric antrum and colon tissue were 0. 662 ±0. 011 and 0. 585 ±0.012 respectively, that were lower than in the control group (P < 0. 01). There was no significant difference in ChAT mRNA expression levels between 6-OHDA group (1.157 ± 0. 037 and 1.154 ± 0. 031) and control group (P > 0.05). Conclusion Constipation and delayed gastric emptying of PD rats may be associated with the increase of dopamine and relative deficiency of cholinergic neurotransmitte in enteric nervous system.
