目的 观察脂多糖(LPS)体外作用于大鼠肝星状细胞(HSC)时肿瘤坏死因子-α(TNF-α)和结缔组织生长因子(CTGF)表达变化,探讨TNF-α对LPS诱导的HSC中CTGF表达的影响.方法 用一步密度梯度离心法分离大鼠HSC,体外培养LPS处理HSC,逆转录-聚合酶链反应(RT-PCR)方法检测不同LPS浓度处理的HSC CTGF mRNA表达水平,并用ELISA双抗体夹心法测定TNF-α浓度.结果 TNF-α水平随LPS浓度增加而增加,LPS可上调CTGF mRNA的表达,TNF-α浓度及CTGF mRNA表达水平呈一定的平行关系.结论 LPS可上调HSC中CTGF mRNA表达,该过程可能是通过TNF-α介导的.

Objective To explore the effect of lipopolysaccharide(LPS)on the expression of connective tissue growth factor(CTGF)in rat hepatic stellate cells(HSC)in vitro and the effect of tumor necrosis factor-α(TNF-α)on the expression of CTGF in HSC induced by LPS.Methods Rat HSCs were separated by in situ perfusion of pronase and eoUengase,and single-step Nyeodenz density gradient centrifugation.After in vitro cultare and passages.HSCs were treated by E coli-derived LPS.The CTGF mRNA expression was detected by simiquantitative RT-PCR with difierent LPS concentrations.TNF-α was determined by enzyme-linked immunosorbnent assay.Results The level of TNF-α was increased with the increased concentrations of LPS,and LPS could also up-regulate the expression of CTGF mRNA and there existed a parallel relationship.Conclusion LPS can up-regulate the expression of CTGF mRNA of HSCs,and TNF-α might play an important role in the pathological process of the expression of CTGF mRNA.
