目的 研究骨髓间充质干细胞(BMSCs)向心肌细胞分化过程中离子通道电流的表达情况,为临床BMSCs移植的安全性提供参考.方法 分离培养BMSCs及乳鼠心肌细胞(NRVMs),并以1: 10的比例共培养,应用全细胞膜片钳技术记录共培养的BMSCs上离子通道电流的表达情况.结果 成功分离培养BMSCs和NRVMs并建立了共培养模型,共培养1周后,BMSCs形态发生了明显变化;在心肌微环境诱导下,向心肌分化的BMSCs平均膜电位为-(40.37±3.68)mV,平均膜电容为(35.18±4.96)pF;共培养的BMSCs上可记录到I_(K1)、I_(to)、I_(Ksus),未记录到I_(Na)和I_(Ca),也未记录到K_(Ca).结论 BMSCs和NRVMs共培养1周后,BMSCs上记录到I_(K1)、I_(to)及I_(Ksus),但未记录到I_(Na)和I_(Ca).

Objective To observe the membrane currents recorded in bone mesenchymal stem cells (BMSCs) differentiating into cardiomyocytes and to provide the reference for the safety and utility of BMSC transplantation. Methods BMSCs and neonatal rat ventricular myocytes (NRVMs) were isolated respectively, and colcultured at a ratio of 1: 10. The whole-cell patch clamp technique was used to record ionic currents in BMSCs after colcultured with cardiomyocytes. Results BMSCs and NRVMs were successfully isolated and cultivated. Then the cocultured model was founded. The average cellular membrane potential of differentiated BMSCs was about -(40.37±3.68)mV, and the mean capacity was about (35.18±4.96)pF. After 1 week coculture, the voltage-clamp experiment showed that I_(K1)、I_(to) and I_(Ksus) were recorded in differentiated BMSCs, but K_(Ca) disappeared. Additionally, I_(Na) and I_(Ca) were still not recorded in differentiated BMSCs. Conclusion I_(K1)、I_(to) and I_(Ksus), but not and I_(Ca) may be recorded in BMSCs differentiating into cardiomyocytes induced by cardiac microenvironment.
