Objective To investigate the characteristics of bacterial L-forms isolated and cultured with blood plates and the causes of the failure detection. Methods 201 specimens were directly inoculated on blood plates to observe their growth. By blind smearing and blind spreading, the pinpoint-shaped and cloudy colonies were taken and their characteristics were observed through the method of dry-up fixation. Those L-forms were identified with routine methods and their biochemical characteristics were determined. Results 102 strains (50.7%) of L-forms were isolated and identified including 57 pinpoint-shaped colonies and 45 cloudy colonies. They were mainly granular form colonies(G-form,81%)and easy to revert(88%). The ultrastructures of L-forms were shown to be deficient in cell walls. Biochemical reactions of L-forms decreased. The L-forms of Staphylococcus aureus ranked first (50%). Conclusions Blood plates are directly used to isolate and culture L-forms. The traditional L-forms characteristics are maintained. It is beneficial to detect the clinical specimens to be bacteria-form and L-forms simultaneously and it can increase the positive rates of detection of L-forms by blood plates.
