以荷叶为材料,在提取温度、提取时间、乙醇体积分数和液料比4个单因素试验的基础上,利用响应曲面试验设计法对荷叶生物碱提取条件进行研究,并通过回归方程和响应曲面,得到的最佳提取工艺为:提取温度84℃,提取时间2.01h,乙醇体积分数90.65%,液料比29.74:1(ml/g).验证实验结果显示,此条件下荷叶生物碱提取率为0.858%.荷叶碱在6.40~32.00μg/ml范围内具有良好的线性关系,r=0.992,荷叶中荷叶碱占荷叶生物碱提取物的比例为7.952%,荷叶中荷叶碱的含量为0.186%.

Optimal condition for alkaloid extraction from lotus leaves was investigated through response surface method based on single factor experiments to study the effect of extraction temper ature, extraction time, alcohol concentration and liquid-material ratio on extraction rate. The optimal extraction condition was 84 ℃ of extraction temperature, 2.01 h of extraction time, 90.65% of alcohol, and 29.74:1 (ml/g) of liquid-material ratio. Results indicated that alkaloid extraction from lotus leaves was 0.858% under this optimal condition. A linear was observed in the range of 6.40-32.00 μg/ml (r = 0.992). The content of nuciferine in alkaloids from lotus leaves was 7.952%.
