Rabbit%Milrinone%Preconditioning%Myocardium protection
目的 探讨米立农预调对成年兔心肌缺血后损伤的保护作用.方法 采用Langendorff离体心脏灌注模型,对32只成年兔随机分为4组进行不同方法预调心肌保护:缺血预调(IPC)、氨力农预调(APC)、米力农预调(MPC),并与单纯全心缺血-再灌注(GI)对照.所有动物均全心缺血30min,开放后再灌注90 min.结果 4组心肌梗死量分别为MPC(10.2±0.7)%、APC(12.8±0.9)%、IPC(23.5±1.5)%和GI(31.2±2.7)%,前两组和后两组比较.差异有统计学意义(P<0.05);前两组左心功能(LVPDP、+dp/dt、LVEDP)恢复亦明显优于后两组(P<0.05).结论 米力农能发挥预调作用,效果与氨力农相当,有可能成为一种新的有效的心肌保护方法.

Objective To investigate the preconditioning effects afforded by milrinone on ischemia reperfusion-induced injury in mature rabbit myocardium. Methods Thirty-two mature rabbit hearts were used for langendorff perfusion. Hearts were initially preconditioning with a exposure to milrinone (MPC),amirinone (APC), ischemia (IPC) or surely global ischemia I.e. Without preconditioning (GI), then all the hearts was subjected to 30-minute ischemia followed by 90-minute reperfusion. Results Infarct size was significantly decreased in MPC and APC as compared with IPC and GI (P<0.05). Left ventricular function (LVPDP, +dp/dt and LVEDP) recovery was also significantly improved in MPC and APC than that in IPC and GI (P<0. 05). Conclusions Milrinone-induced preconditioning makes the heart more tolerant to ischemia-repedusion injury in terms of infarction size and left ventricular function, and as effective as amirinone. This preconditioning may be an ahemative novel and effective myocardial protection strategy.
