观察幼年大鼠发生肺炎时右心室心肌细胞膜L型Ca2+通道电流,研究肺炎合并心力衰竭时心肌细胞钙代谢改变。取50日龄Wistar大鼠,分别经气管注入金黄色葡萄球菌和无菌生理盐水,建立肺炎模型和对照。急性酶解分离出右室心肌细胞,采用全细胞式膜片钳方法,对接种后24h、120h的肺炎组和相应对照组分别记录右室心肌细胞L型Ca2+通道电流。24h肺炎组心肌细胞L型Ca2+通道电流大小无显著改变,而120h肺炎组通道电流显著增大,两组电流曲线的峰电位和激活电位均无改变。实验表明,肺炎早期右室心肌细胞膜L型Ca2+通道电流无明显改变,中后期明显增大,整个观测期间肺炎对该通道基本属性无明显影响。

To probe the myocardial calcium metabolism during heart failure subsequent to acute pneumonia, the L-type Ca2+ current through the right ventricular myocyte membrane of juvenile rat pneumonia models was recored. 50-day old Wister rats were given via trachea staphylococci aureus or sterile normal saline as control. Right ventricular myocytes were enzymatically isolated and L-type Ca2+ currents were recorded with the whole-cell clamp technique. The results showed that the currents were not significantly changed 24 hours after staphylococci aureus administration but were significantly augmented 120 hours after staphylococci aureus administration.No change was found in peak potential and activation potential. It is concluded that L-type Ca2+ currents of right ventricular myocytes don′t change during early stage of acute pneumonia but are augmented during later period. And pneumonia has little effect on peak potential and activation potential.
