CDC20 EGR-1 liver regeneration mitotic progression
目的确认肝再生过程中参与调控肝细胞有丝分裂的EGR1靶向基因CDC20, 并对其作用进行评价.方法通过cDNA微阵列分析,比较切肝后48 h野生鼠和EGR1基因敲出鼠的肝组织基因表达.使用RT-PCR法测定再生肝组织中野生鼠和EGR1基因敲出鼠的CDC20基因表达谱,并将其切肝后48h的结果与同一时间点的EGR1蛋白质表达水平进行比较.结果切肝后前12h肝组织CDC20基因的表达水平无变化,但于36h开始上升并于48h达到顶峰,这个表达曲线恰好与肝再生过程中EGR1蛋白质的表达特点一致.此外,与EGR1基因敲出鼠相比,野生鼠切肝后48h的CDC20基因表达水平明显升高(P<0.02).结论 EGR1基因敲出鼠的肝细胞有丝分裂进程受损与肝再生过程中CDC20基因表达水平降低有关;提示:受EGR1调控的CDC20基因的表达在肝再生过程中肝细胞有丝分裂进程中起重要作用.

Objective:To identify and evaluate the genetic target of EGR-1,CDC20 gene,which involved in regulating hepatocellular mitotic progression during liver regeneration.Methods:Hepatic gene expressions in wildtype and EGR-1 null mouse liver at 48 hours after partial hepatectomy were compared by cDNA microarray.Pattern of CDC20 gene expression were measured by real time RT-PCR in wildtype and EGR-1 null regenerating mouse liver,and the results at 48 hours after partial hepatectomy were compared with our previous EGR-1 protein expression level at this time point.Results:Hepatic CDC20 gene expression was unchanged over the first 12 hours following partial hepatectomy,and then subsequently increased beginning at 36 hours and peaking at 48 hours after partial hepatectomy,which corresponds closely with that of EGR-1 protein expression during liver regeneration.CDC20 was identified as a more highly expressed gene at 48 hours after partial hepatectomy in wildtype regenerating liver compared with EGR-1 null mouse (P<0.02).Conclusion:Impaired mitotic progression is associated with decreased CDC20 expression in regenerating EGR-1 null mouse liver and it suggests that EGR-1 regulation of CDC20 gene expression is an important determinant of hepatocellular mitotic progression during liver regeneration.
