目的 检测胆管癌和癌旁0.5 cm胆管组织及手术切缘的正常胆管组织细胞周期素D1(Cyclin D1)蛋白及mRNA的表达,探讨其在胆管癌发生发展中的作用.方法 应用流式细胞分析术检测42例胆管癌和20例正常胆管黏膜组织Cyclin D1蛋白的表达,同时应用逆转录-聚合酶链反应(RT-PCR)检测42例术中所取的新鲜的胆管癌、17例同个体癌旁胆管黏膜和20例正常胆管组织中Cyclin D1 mRNA表达,并与临床资料进行相关分析.结果 正常胆管组织、癌旁组织、胆管癌组织中Cyclin D1 mRNA相对表达量分别为0.5845±0.0310、0.5608 ±0.0577、0.6288±0.0942,Cyelin D1 mRNA在3组之间表达呈上升趋势(P<0.05).Cyclin D1蛋白表达趋势同其相对应的基因表达趋势一致,即Cyclin D1的mRNA在胆管癌组织中高表达(P<0.05).结论 Cyclin D1基因转录和蛋白可能参与了胆管癌的发生发展过程.
目的 檢測膽管癌和癌徬0.5 cm膽管組織及手術切緣的正常膽管組織細胞週期素D1(Cyclin D1)蛋白及mRNA的錶達,探討其在膽管癌髮生髮展中的作用.方法 應用流式細胞分析術檢測42例膽管癌和20例正常膽管黏膜組織Cyclin D1蛋白的錶達,同時應用逆轉錄-聚閤酶鏈反應(RT-PCR)檢測42例術中所取的新鮮的膽管癌、17例同箇體癌徬膽管黏膜和20例正常膽管組織中Cyclin D1 mRNA錶達,併與臨床資料進行相關分析.結果 正常膽管組織、癌徬組織、膽管癌組織中Cyclin D1 mRNA相對錶達量分彆為0.5845±0.0310、0.5608 ±0.0577、0.6288±0.0942,Cyelin D1 mRNA在3組之間錶達呈上升趨勢(P<0.05).Cyclin D1蛋白錶達趨勢同其相對應的基因錶達趨勢一緻,即Cyclin D1的mRNA在膽管癌組織中高錶達(P<0.05).結論 Cyclin D1基因轉錄和蛋白可能參與瞭膽管癌的髮生髮展過程.

Objective To detect the expression of Cyclin D1 protein and gene in cholangioncarcinoma and to elucidate its role. Methods The expression of Cyclin D1 protein was detected by flow cytometry ( FCM ) in 42 cases of cholangioncarcinoma and 20 cases of normal bile duct, and the expression of Cyclin D1 gene was assayed in 42 cases of cholangioncarcinoma, 17 cases of tissues adjacent to cholangioncarcinoma and 20 cases of normal bile duct by reverse transcription-polymerase chain reaction ( RT-PCR) .The correlation was analyzed statistically between the expression of Cyclin D1 protein and mRNA and clinicopathological parameters in cholangioncarcinoma. Results The expression of Cyclin D1 mRNA in normal bile duct, tissues adjacent to cholangioncarcinoma, and cholangioncarcinoma was 0.5845 ± 0.0310,0. 5608 ± 0. 0577, 0. 6288 ± 0. 0942 respectively. The expression of Cyclin D1 mRNA was higher in cholangioncarcinoma than in others (P <0. 05). The protein expression of Cyclin D1 was higher in cholangioncarcinoma than in normal bile duct tissue (P<0. 05). Conclusion Cyclin D1 gene transcription and protein expression is mostly likely involved in the proliferation and malignant transformation of bile epithelial cells and then malignant progression of bile duct carcinomas. The detection of Cyclin D1 expression may elucidate the carcinogenesis of bile duct.
