目的 观察5-氨基酮戊酸光动力疗法(5-ALA-PDT)作用于人类鼻咽癌细胞株CNE细胞后细胞生长抑制、迁移和侵袭性的改变。方法通过MTT法检测5-ALA-PDT CNE细胞后,不同参数(5-ALA浓度及激光器能量密度)下的细胞生长抑制率变化。并观察不同参数下5-ALA-PDT后细胞划痕实验及Transwell侵袭小室试验。结果5-ALA-PDT能明显抑制鼻咽癌高分化CNE细胞株的生长。5-ALA-PDT后,当药物浓度和能量密度达到一定水平时CNE细胞的迁移被抑制。并且药物浓度在饱和量1 mmol/L时,细胞迁移距离与光照时间呈线性负相关(P<0.05)。光动力作用后,CNE细胞的侵袭能力也被抑制,与5-ALA浓度及激光的能量密度相关。在浓度相同的情况下,细胞侵袭数与照光时间呈线性负相关(r>0.8,P< 0.0001)。结论5-ALA-PDT作用于CNE细胞后,细胞的迁移和侵袭能力被抑制,抑制作用与能量密度和药物浓度相关。

목적 관찰5-안기동무산광동력요법(5-ALA-PDT)작용우인류비인암세포주CNE세포후세포생장억제、천이화침습성적개변。방법통과MTT법검측5-ALA-PDT CNE세포후,불동삼수(5-ALA농도급격광기능량밀도)하적세포생장억제솔변화。병관찰불동삼수하5-ALA-PDT후세포화흔실험급Transwell침습소실시험。결과5-ALA-PDT능명현억제비인암고분화CNE세포주적생장。5-ALA-PDT후,당약물농도화능량밀도체도일정수평시CNE세포적천이피억제。병차약물농도재포화량1 mmol/L시,세포천이거리여광조시간정선성부상관(P<0.05)。광동력작용후,CNE세포적침습능력야피억제,여5-ALA농도급격광적능량밀도상관。재농도상동적정황하,세포침습수여조광시간정선성부상관(r>0.8,P< 0.0001)。결론5-ALA-PDT작용우CNE세포후,세포적천이화침습능력피억제,억제작용여능량밀도화약물농도상관。
Objective To observe and detect the change of invasion and migration about CNE cell line after 5-ALA Photodynamic therapy. Methods The relation between the inhibition rate and 5-ALA concentration & energy density of lasers was detected by MTT. Scratch wound assay and matrigel artificial basement membrane invasion assay were perforned after photodynamic therapy with different photosensitizer concentration and energy density of lasers. Results The results showed that photodynamic therapy can inhibit the growth of CNE cells. The migration of CNE cell was inhibited with appropriate photosensitizer concentration and energy density of laser after photodynamic therapy. It exhibited a linear relation between cell migration and the energy density, when the photosensitizer concentration was a maximum effective concentration of 1 mmol/L (P <0.05). The invasion of CNE cell was also inhibited after photodynamic therapy,there is a relationship between the change of invasion and 5-ALA concentration & energy density of lasers. It which exhibited a linear relation between cell invasion and the energy density in the same photosensitizer concentration (r >0.8, P <0.0001). Conclusion The migration and invasion of CNE cells are both inhibited after photodynamic therapy. The change of migration and invasion is related to 5-ALA concentration & energy density of laser.
