目的 观察大鼠海马脑片盲法全细胞记录技术研究 CA1区锥体神经元电压门控性 Ca2+通道的动力学特征,为在海马脑片上进行神经科新药开发提供依据. 方法选用出生 20~ 30 d的 Wistar雄性大鼠 20只,断头取脑,将海马切为厚 400 μ m的薄片,解剖镜下选 CA1区锥体神经元细胞线行盲法全细胞记录. 结果大鼠海马脑片 CA1区锥体神经元电压门控性 Ca2+通道电流具有如下特点:①激活的阈电位偏低,为(- 49± 9) mV,范围为- 65~- 30 mV( n=23).②衰减时间常数τ值较大,且变化范围大( 100~ 700 ms) (n=12),并且衰减具有 Ca2+电流幅值的依赖性.③稳态失活呈现电压依赖性,半失活电压为(- 55± 10) mV,斜率因子为( 5.3± 0.9) (n=10).④当细胞外 Ca2+浓度为 2.5 mmol/L时, Ca2+通道的反转电位为( 55± 13) mV( n=10).⑤尾电流成分较为单一,不表现电压依赖性.另外, Ca2+电流对戊脉胺及双氢吡啶类化合物硝苯地平均不敏感. 结论海马脑片 CA1区锥体神经元上的 Ca2+通道主要以 N型为主.

목적 관찰대서해마뇌편맹법전세포기록기술연구 CA1구추체신경원전압문공성 Ca2+통도적동역학특정,위재해마뇌편상진행신경과신약개발제공의거. 방법선용출생 20~ 30 d적 Wistar웅성대서 20지,단두취뇌,장해마절위후 400 μ m적박편,해부경하선 CA1구추체신경원세포선행맹법전세포기록. 결과대서해마뇌편 CA1구추체신경원전압문공성 Ca2+통도전류구유여하특점:①격활적역전위편저,위(- 49± 9) mV,범위위- 65~- 30 mV( n=23).②쇠감시간상수τ치교대,차변화범위대( 100~ 700 ms) (n=12),병차쇠감구유 Ca2+전류폭치적의뢰성.③은태실활정현전압의뢰성,반실활전압위(- 55± 10) mV,사솔인자위( 5.3± 0.9) (n=10).④당세포외 Ca2+농도위 2.5 mmol/L시, Ca2+통도적반전전위위( 55± 13) mV( n=10).⑤미전류성분교위단일,불표현전압의뢰성.령외, Ca2+전류대무맥알급쌍경필정류화합물초분지평균불민감. 결론해마뇌편 CA1구추체신경원상적 Ca2+통도주요이 N형위주.
AIM:To investigate the dynamic properties of voltage gated Ca2+ channel of CA1 pyramidal neuron in rat hippocampal slices by using blind whole-cell recording techniques and to provide basis for development of neurological drugs. METHODS:20 Wistar rats(20- 30 d years) were selected.Hippocampus slices of 400 μ m thickness were cut and pyramid neurons at CA1 area were used for blind whole cell recording.RESULTS:The voltage gated Ca2+ channel of CA1 pyramidal neuron in rat hippocampal slices had following features: ① The threshold potential of activation was relatively lower. The mean value was (- 49± 9) mV with range from - 65 to - 30 mV (n=23). ② Current decay was Ca2+ dependent and its time constant (τ ) was longer with a wide range from 100 to 700 ms (n=12). ③ The steady-state inactivation was voltage dependent and the half inactivation potential (V1/2) was (- 55± 10) mV, slope factor (k) was 5.3± 0.9 (n=10). ④ The reversal potential (Erev) of Ca2+ channel was (55± 13) mV (n=10) when extracellular Ca2+-concentration was 2.5 mmol/L.⑤ Tail current, which was voltage independent, primarily had only one component.In addition, Ca2+-channel was insensitive to veripamil and nifedipine. CONCLUSION:N-type Ca2+ channel is the main type in CA1 pyramidal neurons in rat hippocampal slices.
