目的 探讨下丘脑衰老时神经递质的变化及何首乌饮延缓下丘脑衰老的作用机制.方法 D-半乳糖连续腹腔注射致亚急性衰老动物模型,造模后灌胃何首乌饮连续60 d后,大鼠断头取下丘脑.采用ELISA法检测各组大鼠SP,β-EP和GnRH的含量;高效液相色谱法(HPLC)以SHIMADZU-10A 高效液相色谱仪和L-ECD-6A电化学检测器检测下丘脑单胺类神经递质NA,DA,5-HT的含量.结果 同正常对照组相比,模型组大鼠下丘脑GnRH、SP明显增高,β-EP与NA、DA、5-HT含量明显降低,差异有统计学意义(P<0.05).与模型组比较,何首乌饮组下丘脑GnRH、SP明显下降,β-EP与NA、DA、5-HT含量显著升高,呈现一定的剂量依赖性.结论 衰老时大鼠下丘脑正常功能被破坏,何首乌饮通过对神经递质紊乱的调节而延缓了下丘脑的衰老.

Objective To investigate the changes of hypothalamus brain neurotransmitter of aging rats and mechanisms of Heshouwuyin(HSWY) on anti hypothalamus aging.Methods The sub-acutely aging model was continually induced by ip injection of D-galactose in rats. Oral HSWY was administered once daily for 60 days after aging induction.The rats were killed by being amputated by head and the hypothalamus was taken. Use the method of ELISA to detect SP, β-EP and GnRH. The concentration of monamine transmitters including noradrenaline (NA), dopamine compared with the normal group (P<0.05). HSWY dose-dependently decreased the (DA) and 5-hydroxypatmien(5-HT) in hypothalamus were detected by high-performance liquid chromatography (HPLC), with electrochemical detection.Results GnRH and SP of rats in the model group increased significantly, β-EP , NA, DA, 5-HT concentration decreased remarkably determination of GnRH and SP in hypothalamus, and raised the concentration of β-EP NA, DA, 5-HT. Conclusion The normal function of hypothalamus is disturbed in aging rats. HSWY has the effects on preventing and delaying senescence of hypothalamus by regulating the disorder of hypothalamus neurotransmitter.
