[目的]对已知半胱氨酸蛋白酶抑制剂基因编码蛋白的分子量、等电点、信号肽、结构域等进行分析.[方法]在NCBI中检索半胱氨酸蛋白酶抑制剂基因,下载相应的氨基酸序列.采用SMART软件预测结构域,用SingalP程序查找信号肽,用TMHMM程序搜寻预测跨膜区.多序列比对采用CLUSTAL W程序.运用MEGA3.1软件,采用Neighbor-joining 法构建进化树.[结果]多序列比对结果表明半胱氨酸蛋白酶抑制剂基因存在高度保守的QxVxG基序,意味着该基序可能对半胱氨酸蛋白酶抑制剂的抑制活性具有重要意义.系统进化分析可预示半胱氨酸蛋白酶抑制半胱氨酸蛋白酶活性在进化过程中可能也是保守的.[结论]该研究可为半胱氨酸蛋白酶抑制剂抑制半胱氨酸蛋白酶的功能研究方面提供理论参考.

[Objective]The molecular weight, isoelectric point, signal peptide, domain and other properties of the encoding protein of the known cystatin genes were analyzed. [Method]Cystatin genes were searched in NCBI and the related amino acids sequences were downloaded. SMART software was used to predict the domain. SingalP program was used to search signal peptide. TMHMM program was used to search and predict the transmembrane domain. CLUSTAL W program was used to make multiple sequence alignment. Using MEGA3.1 software, the phylogenetic tree was constructed by using Neighbor-joining method. [Result]Multiple sequence alignment showed that cystatin genes had QxVxG base sequences with high conservation, which indicated that the inhibition activity of cystatin had important significance. The phylogenetic analysis might predict that inhibitor activity for cysteine proteinase might be conserved in the evolution process. [Conclusion]The research provided theoretical basis for the research on the inhibition of cystatin on cysteine proteinase.
