目的 观察-1195G>A单核苷酸多态对胰腺癌细胞中COX-2基因启动子转录活性的影响.方法 采用PCR-RFLP对COX-2基因-1195位点各基因型进行分型;构建COX-2基因启动子区pA-1195和pG-1195荧光素酶报告基因表达质粒;转染胰腺癌细胞系Asp-1;采用双荧光素酶报告基因方法,检测-1195AA和-1195GG基因型对报告基因表达的影响.结果 与含-1195GG基因型的COX-2启动子相比,含-1195AA基因型的COX-2启动子,在胰腺癌细胞Aspc-1具有较高的报告基因表达水平.重组质粒pA1195驱动的报告基因表达水平为pG1195驱动的报告基因表达水平的17.83倍.结论 COX-2基因启动子区的-1195G>A单核苷酸多态-1195位点G→A改变明显增加胰腺癌细胞COX-2基因启动子的转录活性.

Objective To investigate the effects of single nucleotide polymorphism of-1195G.>A in the promoter of COX-2 gene on the transcriptional activity of COX-2 gene in the pancreatic cancer cell line. Methods The genotypes of-1195AA and-1195GG of COX-2 gene were deter-mined by polymerase chain reaction-based restriction fragment length polymorphism (PCR-RFLP).The reporter plasmids with-1195AA and-1195GG genotypes were constructed and then the pan-creatic Aspc-lcancer cells were transfected with these two kinds of plasmaids. The transcriptional ac-tivity of the COX-2 promoter in Aspc-1 transfected with-1195AA or-1195GG genotype was tested by luciferase report gene assay. Results The transcriptional activity of the COX-2 promoter in Aspc-1 transfected with-1195AA genotype was as 17.83 times higher as that with-1195GG genotype.Conclusion The single nucleotide polymorphism of-1195G.>A can greatly up-regulate the tran-scriptional activity of the promoter of COX-2 gene in the pancreatic cancer cells.
