[目的]提高大肠杆菌DH5α转化效率.[方法]在其他条件一致的情况下,将5 μl的质粒或连接产物加入含有100 μl 的感受态0.2 ml PCR管和1.5 ml的离心管中,0.2 ml PCR管设置热休克时间梯度和1.5 ml的离心管热休克时间90 s,对大肠杆菌DH5α转化效率进行了比较.[结果]0.2 ml PCR管转化效率高于1.5 ml的离心管,且0.2 ml PCR管热休克30 s时最高.[结论]采用此方法提高了大肠杆菌DH5α的转化效率.
[目的]提高大腸桿菌DH5α轉化效率.[方法]在其他條件一緻的情況下,將5 μl的質粒或連接產物加入含有100 μl 的感受態0.2 ml PCR管和1.5 ml的離心管中,0.2 ml PCR管設置熱休剋時間梯度和1.5 ml的離心管熱休剋時間90 s,對大腸桿菌DH5α轉化效率進行瞭比較.[結果]0.2 ml PCR管轉化效率高于1.5 ml的離心管,且0.2 ml PCR管熱休剋30 s時最高.[結論]採用此方法提高瞭大腸桿菌DH5α的轉化效率.

[Objective]The research aimed to improve the transformation efficiency of E.coli DH5α.[Method]Under the same other conditions, 5 μl plasmid or legation product was added into 0.2 ml PCR tube and 1.5 ml centrifuge tube with 100 μl competent in them. In different thermal shock time, transformation efficiency of E.coli DH5α was compared.[Result]Transformation efficiency with 0.2 ml PRC reformer tube is better than that with 1.5 ml centrifuge tube and the 0.2 ml PCR reformer tube with 30 seconds' thermal shock time gives the best performance of transformation efficiency.[Conclusion]Transformation efficiency of E.coli DH5α is effectively improved by this method.
