目的:对体外诱导小鼠脾脏单个核细胞分化成的树突状细胞(DC)的生物学表型及功能进行检测,并与骨髓来源的DC进行比较.方法:分离小鼠脾脏单个核细胞,在体外培养的条件下通过添加25 ng·ml-1的粒-单核细胞集落刺激因子和25 ng·ml-1的白细胞介素-4(IL-4)将其诱导为DC,分别从细胞形态、表型以及功能3个方面对其进行检测,并与骨髓来源的DC进行比较.结果:小鼠脾脏来源的DC经磷酸脂多糖(LPS)刺激成熟后其表面显示出明显的树枝状突起,高表达CD11c、CD86及MHC-Ⅱ类分子,并具有极强的刺激同种异基因淋巴细胞增殖的能力,其特征与骨髓来源的DC相差无几.结论:小鼠脾脏单个核细胞能够被诱导为具有正常形态、表型、功能的DC,为DC的功能研究以及进一步制备相应的肿瘤疫苗提供又一可靠来源.
目的:對體外誘導小鼠脾髒單箇覈細胞分化成的樹突狀細胞(DC)的生物學錶型及功能進行檢測,併與骨髓來源的DC進行比較.方法:分離小鼠脾髒單箇覈細胞,在體外培養的條件下通過添加25 ng·ml-1的粒-單覈細胞集落刺激因子和25 ng·ml-1的白細胞介素-4(IL-4)將其誘導為DC,分彆從細胞形態、錶型以及功能3箇方麵對其進行檢測,併與骨髓來源的DC進行比較.結果:小鼠脾髒來源的DC經燐痠脂多糖(LPS)刺激成熟後其錶麵顯示齣明顯的樹枝狀突起,高錶達CD11c、CD86及MHC-Ⅱ類分子,併具有極彊的刺激同種異基因淋巴細胞增殖的能力,其特徵與骨髓來源的DC相差無幾.結論:小鼠脾髒單箇覈細胞能夠被誘導為具有正常形態、錶型、功能的DC,為DC的功能研究以及進一步製備相應的腫瘤疫苗提供又一可靠來源.
목적:대체외유도소서비장단개핵세포분화성적수돌상세포(DC)적생물학표형급공능진행검측,병여골수래원적DC진행비교.방법:분리소서비장단개핵세포,재체외배양적조건하통과첨가25 ng·ml-1적립-단핵세포집락자격인자화25 ng·ml-1적백세포개소-4(IL-4)장기유도위DC,분별종세포형태、표형이급공능3개방면대기진행검측,병여골수래원적DC진행비교.결과:소서비장래원적DC경린산지다당(LPS)자격성숙후기표면현시출명현적수지상돌기,고표체CD11c、CD86급MHC-Ⅱ류분자,병구유겁강적자격동충이기인림파세포증식적능력,기특정여골수래원적DC상차무궤.결론:소서비장단개핵세포능구피유도위구유정상형태、표형、공능적DC,위DC적공능연구이급진일보제비상응적종류역묘제공우일가고래원.
Objective: To induce dendritic cells from mouse spleen mononuclear cells in vitro, and to examine the morphology, phenotype and function of the dendritic cells. Methods: Mononuclear cells isolated from mouse spleen cells were induced into dendritic cells in vitro by morphology, phenotype and function were examined. Results: Mouse spleen derived DCs were induced into mature through exposure to lipopolysaccharide(LPS) and exhibited dramatic veils of cytoplasm and extensive dendrites in their surfaces with high expression of CD11c, MHC-Ⅱ and CD86. Their capacity to stimulate primary T cell responses in mixed lymphocyte reaction(MLR) increased. The characteristics of mouse spleen derived DCs were similar to those of bone marrow DCs(BM-DCs). Conclusion: Mouse spleen derived mononuclear cells could be induced into DCs with normal morphology, phenotype and function, which provides a new reliable source for the research of DC function and the preparation of corresponding anti-tumor vaccine.
