本研究采用DAS-ELISA、IC-RT-PCR及序列测定方法,对加拿大进境大豆种子进行菜豆荚斑驳病毒(Bean pod mottle virus,BPMV)和大豆花叶病毒(Soybean mosaic virus,SMV)检测,结果表明,BPMV的DAS-ELISA和IC-RT-PCR检测结果均为阳性,且PCR扩增产物序列与已报道的BPMV基因序列相似性达97%以上,而SMV的DAS-ELISA和IC-RT-PCR检测结果都为阴性.综合血清学、分子生物学检测结果,确认该批大豆携带有菜豆荚斑驳病毒.

본연구채용DAS-ELISA、IC-RT-PCR급서렬측정방법,대가나대진경대두충자진행채두협반박병독(Bean pod mottle virus,BPMV)화대두화협병독(Soybean mosaic virus,SMV)검측,결과표명,BPMV적DAS-ELISA화IC-RT-PCR검측결과균위양성,차PCR확증산물서렬여이보도적BPMV기인서렬상사성체97%이상,이SMV적DAS-ELISA화IC-RT-PCR검측결과도위음성.종합혈청학、분자생물학검측결과,학인해비대두휴대유채두협반박병독.
The presence of BPMV and SMV in soybean seeds imported from Camda was determined using DASELISA,IC-RT-PCR and sequence analysis.The results showed that both DAS-ELISA and IC-RT-PCR were positive for BPMV,and the nucleotide sequence of BPMV PCR product had more than 97% similarity to previously reported genornic sequence.However,negative DAS-ELISA and IC-RT-PCR results were obtained for SMV.According to the results of serological and molecular biological methods,it was confirmed that BPMV was present in tested soybean imported from Camda.
