以大肠杆菌为工程菌,针对β-胡萝卜素的累积和提取两大方面,研究了不同IPTG诱导时间及浓度、菌液OD值、盐酸用量、破壁时间及温度、丙酮用量、抽提时间8个条件对β-胡萝卜素累积及提取的影响,并用分光光度计测定不同条件下工程菌积累的β-胡萝卜素的吸光度值,从而筛选适合β-胡萝卜素高效累积及提取的最佳培养条件.结果表明,当诱导时间为1.25 h,IPTG浓度为0.8 mmol/L,菌液OD值为0.6,HCl量为15 mL/g(湿菌体),破壁时间为40 min,破壁温度为40℃,丙酮量为20 mL/g(湿菌体),抽提时间为0.5h时,β-胡萝卜素的提取量达到最大.
以大腸桿菌為工程菌,針對β-鬍蘿蔔素的纍積和提取兩大方麵,研究瞭不同IPTG誘導時間及濃度、菌液OD值、鹽痠用量、破壁時間及溫度、丙酮用量、抽提時間8箇條件對β-鬍蘿蔔素纍積及提取的影響,併用分光光度計測定不同條件下工程菌積纍的β-鬍蘿蔔素的吸光度值,從而篩選適閤β-鬍蘿蔔素高效纍積及提取的最佳培養條件.結果錶明,噹誘導時間為1.25 h,IPTG濃度為0.8 mmol/L,菌液OD值為0.6,HCl量為15 mL/g(濕菌體),破壁時間為40 min,破壁溫度為40℃,丙酮量為20 mL/g(濕菌體),抽提時間為0.5h時,β-鬍蘿蔔素的提取量達到最大.
이대장간균위공정균,침대β-호라복소적루적화제취량대방면,연구료불동IPTG유도시간급농도、균액OD치、염산용량、파벽시간급온도、병동용량、추제시간8개조건대β-호라복소루적급제취적영향,병용분광광도계측정불동조건하공정균적루적β-호라복소적흡광도치,종이사선괄합β-호라복소고효루적급제취적최가배양조건.결과표명,당유도시간위1.25 h,IPTG농도위0.8 mmol/L,균액OD치위0.6,HCl량위15 mL/g(습균체),파벽시간위40 min,파벽온도위40℃,병동량위20 mL/g(습균체),추제시간위0.5h시,β-호라복소적제취량체도최대.
In this paper, we studied the effects of different IPTG induction time and concentration, the OD values of engineering strain (E coli), hydrochloric acid dosage, broken time and temperature, acetone dosage and time of extraction on the accumulation and extraction of β -carotene to select the best accumulation conditions of β-carotene. The results showed that: when the IPTG induction time was 1.25 h, IPTG concentration was 0.8 mmol/L, OD value of E cali was 0.6, HCl volume per wet pellet was 15 mL/g, HCl broken time was 40 min, HCl broken temperature was 40 ℃, acetone volume was 20 mL/g wet biomass, and acetone extraction time is 0.5 h, the accumulation of β -carotene was up to maximum.
