借助表皮条实验和激光扫描共聚焦显微镜技术,研究了天然和合成生长素IAA、NAA、2,4-D、细胞分裂素ZT、KT、6-BA在光、暗调控气孔运动中的作用及其机制.结果表明,暗中所试生长素、细胞分裂素、NO清除剂cPTIO、Hb、NO合酶抑制剂L-NAME、H_2O_2清除剂AsA、CAT、H_2O_2产生酶NADPH氧化酶抑制剂DPI对气孔开放的效应显著大于光下,暗示内源生长素、细胞分裂素、NO、H_2O_2均参与光/暗调控气孔运动,光下内源生长素、细胞分裂素浓度大于暗中,但暗中内源NO、H_2O_2水平高于光下.另外,所试生长素、细胞分裂素和NO、H_2O_2清除剂及合成抑制剂对光、暗条件下气孔运动效应的一致性还表明,生长素、细胞分裂素可能通过降低保卫细胞内源NO、H_2O_2促进气孔开放,内源NO、H_2O_2检测的结果正好证明了这一推论.

The roles and mechanisms of natural and synthetic auxin IAA, NAA, 2,4-D, cytokinin ZT, KT, 6-BA in light/dark-regulated stomatal movement were investigated by means of stomatal bioassay and using laser-scanning congocal microscopy. The results showed that the effects of auxin IAA, NAA, 2,4-D, cytokinin ZT, KT, 6-BA and NO scavenger cPTIO, Hb, NOS inhibitor L-NAME, H_2O_2 scavenger AsA, CAT, NADPH oxidase inhibitor DPI on stomatal opening were more significant in dark than in light, suggesting that endogenous auxin, cytokinin, NO and H_2O_2 mediated light/dark-regulated stomatal movement, and the levels of endogenous auxin and cytokinin were more higher in light than in dark. However, The results indicated that endogenous NO and H_2O_2 were more greater in dark than in light. In addition, the effects of auxin and cytokinin tested on stomatal movement in light or in darkness are accordant with that of NO and H_2O_2 scavengers, synthetic inhibitors, the results imply that auxin and cytokinin probably promoted stomatal opening via decreasing endogenous NO and H_2O_2, which was verified by the monitoring results of endogenous NO, H_2O_2 fluorescence.
