Krüppel样锌指因子是真核生物中广泛存在的一类转录调控因子,参与细胞分化、增殖、凋亡和肿瘤形成等多种生理病理过程.其结构特点是羧基端高度保守,含有3个C2H2型锌指结构,与富含GC盒或CACCC序列的DNA结合;而氨基端的转录调控区在不同因子之间差异较大,发挥转录激活或转录抑制效应.现有研究显示多个Krüppel样锌指因子成员参与调控各类血细胞的生命活动.此文将就这几个与血液疾病发生有关的KLF样锌指因子作一概述.
Krüppel樣鋅指因子是真覈生物中廣汎存在的一類轉錄調控因子,參與細胞分化、增殖、凋亡和腫瘤形成等多種生理病理過程.其結構特點是羧基耑高度保守,含有3箇C2H2型鋅指結構,與富含GC盒或CACCC序列的DNA結閤;而氨基耑的轉錄調控區在不同因子之間差異較大,髮揮轉錄激活或轉錄抑製效應.現有研究顯示多箇Krüppel樣鋅指因子成員參與調控各類血細胞的生命活動.此文將就這幾箇與血液疾病髮生有關的KLF樣鋅指因子作一概述.

Krüppel-like factors are an important component of the transcription machinery in eukaryotic cells,which are involved in many physiologic and pathologic processes,including cell proliferation,differentiation,apoptosis and neoplasm transformation.They have a highly conserved domain at carboxyl terminal that binds to GC-rich regions or CACCC sequence in DNA via three C2H2 zinc motifs,whereas their amino terminal is vary greatly and contains transcription regulatory domains to mediate transcriptional activation or repression.Recently,several groups have been identified to control the diverse biological processes of erythrocytes and leukocytes.For example,KLF1 activates β-globin gene expression;KLF2 has been demonstrated to regulate T-cell properties of quiescence and survival ; KLF4,KLF5 and KLF6 are involved in the pathogenesis of blood cell cancers. In this article,we focuses on those KLFs which have new connection with blood disease.
