试验成功地培养了猪颗粒细胞,并采用激光共聚焦显微技术研究细胞有丝分裂过程中Ran的定位变化.结果表明,猪间期颗粒细胞中,Ran主要分布于细胞核内,核仁内没有Ran分布,细胞质中Ran浓度极低.核膜破裂,细胞进入有丝分裂,Ran分布到整个细胞,但在染色体周围浓集;在有丝分裂中期,浓集区形态和纺锤体的形态一致;在有丝分裂后期和末期,Ran的浓集区随纺锤体的拉长而分布于被分开的两组染色体之间;有丝分裂结束后,随着细胞核的形成,Ran在染色体周围浓集,核膜形成后又浓集于细胞核内,在整个有丝分裂中,Ran在细胞分裂区的细胞膜下未见特殊分布.这些分布与Ran在细胞周期中的作用是一致的.
試驗成功地培養瞭豬顆粒細胞,併採用激光共聚焦顯微技術研究細胞有絲分裂過程中Ran的定位變化.結果錶明,豬間期顆粒細胞中,Ran主要分佈于細胞覈內,覈仁內沒有Ran分佈,細胞質中Ran濃度極低.覈膜破裂,細胞進入有絲分裂,Ran分佈到整箇細胞,但在染色體週圍濃集;在有絲分裂中期,濃集區形態和紡錘體的形態一緻;在有絲分裂後期和末期,Ran的濃集區隨紡錘體的拉長而分佈于被分開的兩組染色體之間;有絲分裂結束後,隨著細胞覈的形成,Ran在染色體週圍濃集,覈膜形成後又濃集于細胞覈內,在整箇有絲分裂中,Ran在細胞分裂區的細胞膜下未見特殊分佈.這些分佈與Ran在細胞週期中的作用是一緻的.

Pig granular cells were successfully cultured and the localization and possible roles of Ran were investigated during the cell mitotic division by using the confocal laser scanning microscopy. The results showed that Ran was localized mainly in the nucleus, except the nucleolus, in the intermediate stage pig granular cell, whose density was very low in the cytoplasm. After the nuclear membrane broke down in mitochysis, Ran distributed throughout the cell, but it concentrated to the chromosome in mitosis metaphase, the concentrated area was uniformity to the spindle; in mitosis anaphase and telephase, the concentrated area localized between the separated chromosomes, but lower in other places. After mitosis, Ran coecentrated around the chromosome, and migrated into the forming nucleus. There is no special distribution for Ran under the membrane of the cell fission. These distributions were consistent with the roles of Ran in the cell cycle.
