本文研究了基本培养基和植物生长调节剂对金钗石斛愈伤组织诱导和增殖的影响,并绘制了生长曲线。结果表明,改良MS2(K2HPO.加倍)能获得60%的诱导率,而改良MS。CNH4NO,加倍)的诱导率仅为5%。二者在愈伤组织培养上也表现出显著差异,前者表现良好,说明高浓度的铵能抑制金钗石斛愈伤组织诱导和增殖。MS+0.5mg/LBA+O.8Ing/LNAA+3%蔗糖可获得最高诱导率和增殖倍数,诱导率为70.0%。90d后增殖倍数为23.98。类原球茎培养60~100d时进入对数生长期,重量快速增加。

The influence of basal medium and plant growth regulator on callus induction of Dcndrobium nobile Lindl was studied and to draft the growth curve of callus in this paper. The result showed that the rate of callus induction had reached 60% on the modified MSI media which double amount of KzHPO 4, as well as only 5%on the modified MS2 media which double amount ofNH 4NO 3:. There were signii'icant difference on callus multiplication between them, the first one was better. This indicates that the callus induction and multiplication of Denclrobium nobile Lindl arc inhibited by high concentration of ammonium. The highest induction rate and proliferation times was got on MS medium plused with 05 mg/L BA, 08 mg/L NAA and 3% sugar. The induction rate was 70%, the proliferation times was 23 98 after 90 days. The weight of callus was multiply and got in the logarithmic phase when culture time from 60 to100 days.
