目的 探讨AKT1与AKT2在脑胶质瘤中的表达及与患者生存期的相关性.方法 免疫荧光法检测手术切除的50例胶质瘤组织标本中AKT1及AKT2的表达水平,确定两者表达的相关性并与生存期进行相关性分析.结果 Log - rank单因素检验,年龄≥50岁、KPS评分< 80分和WHO Ⅲ~Ⅳ级患者的生存期显著缩短.胶质瘤组织中AKT1及AKT2在WHOⅢ~Ⅳ级的表达率显著高于WHO Ⅰ~Ⅱ级,两者表达存在正相关,且与患者的生存期呈负相关.结论 AKT1与AKT2的表达状态对预测患者的生存期具有一定的临床价值.

Objective To investigate the expression of AKT1 and AKT2 and its correlation with the overall survive time of glioma patients.Method Glioma patients treated with resection from January 2006 to August 2006 in our hospital were analyzed retrospectively.Immunofluorescence staining was applied to detect the expression of AKT1 and AKT2 in 50 tissue samples of glioma patients with integral following information.The relationship of AKT1 and AKT2 expression with the overall survival time and the correlation between two proteins were analyzed.Results Log- rank test revealed that age ≥ 50 years,preoperative KPS score < 80 and WHO Grade Ⅲ ~ Ⅳ were statistically significant factors ( P < 0.05) for overall survival time in the univariate analysis.The positive rates of AKT1 and AKT2 expression in glioma patients were separately 80%,82%.The levels of AKT1 and AKT2 protein increased markedly in high grade gliomas in comparison to low grade gliomas.The positive rates of two proteins expression of WHO Grade Ⅲ ~ Ⅳ and WHO Grade Ⅰ ~ Ⅱ were separately 94%,94% ;43%,50%.There was a positive linear correlation between the AKT1 and AKT2 expression( r =0.8761,P < 0.0001 )and a negative correlation was found between the expression of two proteins and the overall survival time of glioma patients ( r =-0.7620,P < 0.0001; r =- 0.8073,P < 0.0001 ).Conclusions Detection of AKT1 and AKT2 is of great value for predicting the biological behaviors of glioma and the overall survival time of glioma patients.
