目的 探讨二烯丙基三硫化物(DAIS)对肝移植术后大鼠肝癌皮下移植瘤生长的影响及其机制.方法 建立大鼠肝移植术后肝癌皮下移植瘤模型24只,随机分为对照组、DAIS Ⅰ组(每日1 mg/kg)、DAISⅡ组(每日15 mg/kg).用药2周后观察肿瘤体积和重量,生化法测定大鼠血清肝肾功能,荧光定量聚合酶链反应(PCR)法检测肿瘤的血管内皮生长因子(VEGF)、基质金属蛋白酶(MMP)-2 mRNA的表达,肿瘤组织切片行透射电镜检查.结果 与对照组比较,DAIS能抑制肿瘤生长,对照组、DAIS Ⅰ组和DAIS Ⅱ组皮下移植瘤体积分别为(0.78±0.25)、(0.42±0.17)、(0.38±0.21)cm3(P<0.01);DAIS抑制肝癌细胞VEGF mRNA、MMP-2 mRNA的转录(P<0.01).DAIS对肝移植术后大鼠肝、肾功能无明显毒性作用(P>0.05);透射电镜下可见DAIS组肿瘤组织内有较多细胞凋亡现象.结论 DAIS可以抑制肝癌细胞VEGFmRNA和MMP-2 mRNA的转录,诱导肿瘤细胞凋亡,抑制肝移植术后皮下移植瘤的生长.

목적 탐토이희병기삼류화물(DAIS)대간이식술후대서간암피하이식류생장적영향급기궤제.방법 건립대서간이식술후간암피하이식류모형24지,수궤분위대조조、DAIS Ⅰ조(매일1 mg/kg)、DAISⅡ조(매일15 mg/kg).용약2주후관찰종류체적화중량,생화법측정대서혈청간신공능,형광정량취합매련반응(PCR)법검측종류적혈관내피생장인자(VEGF)、기질금속단백매(MMP)-2 mRNA적표체,종류조직절편행투사전경검사.결과 여대조조비교,DAIS능억제종류생장,대조조、DAIS Ⅰ조화DAIS Ⅱ조피하이식류체적분별위(0.78±0.25)、(0.42±0.17)、(0.38±0.21)cm3(P<0.01);DAIS억제간암세포VEGF mRNA、MMP-2 mRNA적전록(P<0.01).DAIS대간이식술후대서간、신공능무명현독성작용(P>0.05);투사전경하가견DAIS조종류조직내유교다세포조망현상.결론 DAIS가이억제간암세포VEGFmRNA화MMP-2 mRNA적전록,유도종류세포조망,억제간이식술후피하이식류적생장.
Objective To study the effect of diallyl trisulfide (DATS) on the growth of hepato-cellular carcinoma(HCC)after orthotopie liver transplantation(OLTx)in rats. Methods Twenty-four rats bearing subcutaneous xenograft HCC after OLTx were randomly divided into 3 groups: the control group,the DATS group Ⅰ(DATS 1 mg/kg every day) and DATS group Ⅱ(DATS 5 mg/kg every day). The effects of DATS on the growth of the tumor were evaluated after 2 weeks,including the tumor size and weight. Fluorescent quantitation ploymerase chain reaction was used to detect the expression of vascular en-dothelial growth factor (VEGF) mRNA and matrix metalloproteinase 2(MMP-2)mRNA. The effects of DATS on the liver and kidney in rats were assayed by biochemical methods. Results DATS inhibited the growth of HCC in rats after OLTx. Tumor size : the control group(0. 78±0.25)cm3, DATS group Ⅰ (0.42±0.17) cm3 ,and DATS group Ⅱ (0.38±0.21)cm3 (P<0.01). The expression of VEGF mR- NA and MMP-2 mRNA was downregulated significantly by DATS (P < 0. 01 ). DATS had no side effects on the liver and kidney in rats(P > 0.05 ). Conclusion DATS can inhibit the growth of HCC in rats af-ter OLTx without side effects in vivo by suppressing the expression of VEGF mRNA and MMP-2 mRNA inHCC.
