目的 探讨中药菊花决明散对大鼠高危角膜移植排斥反应的抑制作用.方法 角膜缝线法诱导受体角膜新生血管,行穿透性角膜移植,建立大鼠高危角膜移植排斥反应模型.术后治疗组采用每天菊花决明散方0.1g/2 mL灌胃,对照组采用生理盐水2 mL灌胃,观察植片存活状况;应用组织病理学、免疫荧光组织化学,流式细胞仪等方法对受体局部和全身免疫状态进行检测.结果 治疗组大鼠角膜植片存活时间为(14.50±3.55)天,明显长于生理盐水组的(8.25±0.71)天(P<0.01);治疗组术后虹膜铺片Fas、FasL的表达低于对照组;术后14天外周血中CD_4~+CD_(25)~+FOXP3~+阳性细胞治疗组[(14.81±2.58)%]高于对照组[(5.11±3.92)%],差异有统计学意义(P<0.01);治疗组房水中IL-2浓度低于对照组而IL-10浓度高于对照组.结论 菊花决明散方对高危角膜移植排斥反应模型大鼠术后免疫排斥反应具有一定抑制作用.
目的 探討中藥菊花決明散對大鼠高危角膜移植排斥反應的抑製作用.方法 角膜縫線法誘導受體角膜新生血管,行穿透性角膜移植,建立大鼠高危角膜移植排斥反應模型.術後治療組採用每天菊花決明散方0.1g/2 mL灌胃,對照組採用生理鹽水2 mL灌胃,觀察植片存活狀況;應用組織病理學、免疫熒光組織化學,流式細胞儀等方法對受體跼部和全身免疫狀態進行檢測.結果 治療組大鼠角膜植片存活時間為(14.50±3.55)天,明顯長于生理鹽水組的(8.25±0.71)天(P<0.01);治療組術後虹膜鋪片Fas、FasL的錶達低于對照組;術後14天外週血中CD_4~+CD_(25)~+FOXP3~+暘性細胞治療組[(14.81±2.58)%]高于對照組[(5.11±3.92)%],差異有統計學意義(P<0.01);治療組房水中IL-2濃度低于對照組而IL-10濃度高于對照組.結論 菊花決明散方對高危角膜移植排斥反應模型大鼠術後免疫排斥反應具有一定抑製作用.
목적 탐토중약국화결명산대대서고위각막이식배척반응적억제작용.방법 각막봉선법유도수체각막신생혈관,행천투성각막이식,건립대서고위각막이식배척반응모형.술후치료조채용매천국화결명산방0.1g/2 mL관위,대조조채용생리염수2 mL관위,관찰식편존활상황;응용조직병이학、면역형광조직화학,류식세포의등방법대수체국부화전신면역상태진행검측.결과 치료조대서각막식편존활시간위(14.50±3.55)천,명현장우생리염수조적(8.25±0.71)천(P<0.01);치료조술후홍막포편Fas、FasL적표체저우대조조;술후14천외주혈중CD_4~+CD_(25)~+FOXP3~+양성세포치료조[(14.81±2.58)%]고우대조조[(5.11±3.92)%],차이유통계학의의(P<0.01);치료조방수중IL-2농도저우대조조이IL-10농도고우대조조.결론 국화결명산방대고위각막이식배척반응모형대서술후면역배척반응구유일정억제작용.
Objective To study the inhibitory effect of Juhua Jueming powder (JJP) on high-risk corneal transplantation immune rejection in rats. Methods The high-risk corneal transplantation immune rejection rat model was established by inducing coneal neoangiogenesis by suture method and the penetrating transplantation. Model rats were divided into two groups,the treated group and the control group,they were administered with JJP 0.1 g (dissolved in 2 mL water) and normal saline respectively via gastric infusion every day after transplantation. The survival status of the allograft,histopathology,local and systemic immune status in the recipients were observed using immunofluorescence histochemistry and flow cytometry. Results The survival time of the allograft in the treated group (14.50±3.55 days) was significantly longer than that in the control group (8.25±0.71 days,P<0.01). Levels of Fas,FasL expressions in iris were stronger,and the percentage of CD_4~+ CD_(25)~+ FOXP3 positive cells in peripheral blood was less (5.11±3.92% vs 14.81±2.58%) in the control group than those in the treated group respectively. The concentration of IL-2 was lower while that of IL-10 was higher in aqueous humor of the treated group than those of the control group,respectively. Conclusion JJP has certain effect for preventing high-risk corneal transplantation immune rejection in rat model.
