本试验旨在研究褪黑素、谷胱甘肽两种抗氧化剂对精子冷冻的保护效应。以成年杜洛克公猪为研究对象,在猪精液冷冻稀释液中先后单独、联合添加褪黑素、谷胱甘肽,解冻后精子质量通过检测活率、活力、顶体完整性、线粒体活性以及活性氧(ROS)含量来判定。结果表明:分别单独添加0.25mg/mL褪黑素、5mmol/L谷胱甘肽,或联合添加0.125mg/mL褪黑素和1mmol/L谷胱甘肽,均能减少精液冷冻过程中ROS的生成,显著提高冷冻精子解冻后的质量(P〈0.05),其中联合添加效果显著优于单独添加效果。

본시험지재연구퇴흑소、곡광감태량충항양화제대정자냉동적보호효응。이성년두락극공저위연구대상,재저정액냉동희석액중선후단독、연합첨가퇴흑소、곡광감태,해동후정자질량통과검측활솔、활력、정체완정성、선립체활성이급활성양(ROS)함량래판정。결과표명:분별단독첨가0.25mg/mL퇴흑소、5mmol/L곡광감태,혹연합첨가0.125mg/mL퇴흑소화1mmol/L곡광감태,균능감소정액냉동과정중ROS적생성,현저제고냉동정자해동후적질량(P〈0.05),기중연합첨가효과현저우우단독첨가효과。
In order to study the effect of melatonin and reduced glutathione on the cryopreservation of boar semen, we added these two antioxidants independently and jointly to the boar semen extenders in succession, then detected the content Of ROS, viability, motility, acrosomal integrity, and mitoehondrial activity of post-thaw se.men. The result showed that added melatonin 0.25 mg/mL, reduced glutathione 5 mmol/L independently, or added melatonin 0.125 mg/mL and reduced glutathione 1 mmoUL jointly could reduce the content of ROS, and improved the quality of post-thaw semen significantly (P〈0.05), and it was better to add iointlv than independently.
