目的 观察Survivin反义核酸载体对人骨肉瘤细胞株MG-63生物学行为的抑制效应,并探讨其机制.方法 MTY检测转染前后MG-63细胞增殖抑制率的变化,Boyden小室体外侵袭实验测定转染前后侵袭能力变化.逆转录-聚合酶链反应(RT-PCR)检测转染前后Ang-2基因mRNA表达的变化.结果 噻唑蓝(MTT)检测显示转染后骨肉瘤细胞增殖明显受抑制,转染组抑制率达到23.4%,与各对照组之间差异有统计学意义(P<0.01),脂质体组、空质粒组和空白组之间差异无统计学意义(P>0.05).RT-PCR分析显示转染组MG-63中Ang-2基因mRNA的表达较转染前明显下降,与其他组之间差异有统计学意义(P<0.01),而另3组之间差异无统计学意义(P>0.05).Boyden小室体外侵袭实验测定显示转染组侵袭百分数为17.9%,与其他各组间差异有统计学意义(P<0.01).结论 Survivin反义核酸可以显著抑制人骨肉瘤细胞株MG-63的增殖、侵袭能力,部分逆转其恶性表型,这可能与其下调Ang-2的表达有关.

목적 관찰Survivin반의핵산재체대인골육류세포주MG-63생물학행위적억제효응,병탐토기궤제.방법 MTY검측전염전후MG-63세포증식억제솔적변화,Boyden소실체외침습실험측정전염전후침습능력변화.역전록-취합매련반응(RT-PCR)검측전염전후Ang-2기인mRNA표체적변화.결과 새서람(MTT)검측현시전염후골육류세포증식명현수억제,전염조억제솔체도23.4%,여각대조조지간차이유통계학의의(P<0.01),지질체조、공질립조화공백조지간차이무통계학의의(P>0.05).RT-PCR분석현시전염조MG-63중Ang-2기인mRNA적표체교전염전명현하강,여기타조지간차이유통계학의의(P<0.01),이령3조지간차이무통계학의의(P>0.05).Boyden소실체외침습실험측정현시전염조침습백분수위17.9%,여기타각조간차이유통계학의의(P<0.01).결론 Survivin반의핵산가이현저억제인골육류세포주MG-63적증식、침습능력,부분역전기악성표형,저가능여기하조Ang-2적표체유관.
Objective To observe the inhibitory effects of an antisense Survivin vector on proliferation and invasion of osteosarcoma cells line MG-63, and explore the mechanism. Methods MTT assay was used to measure the proliferation inhibition after transfection of MG-63 cells with Survivin antisense RNA. Boyden chamber in vitro invasion assay was applied to observe the changes of invasion capability.The expression of Angd-2 before and after transfection was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The proliferation of cells transfected with antisense Survivin was significantly inhibited ( the inhibition rate was 23.4% ) as compared with other groups. The invasion ability was remarkably decreased after antisense transfection ( 17. 9% ) as compared with the control groups. Moreover,the mRNA expression of Ang-2 in antisense Survivin transfected cells was much lower than control groups.Conclusion The antisense Survivin vector could markedly inhibit the proliferation and invasion of osteosarcoma cells line MG-63, and partially reversed their malignant phenotype, which was possibly related with down-regulation of the Ang-2 expression.
