以14个黑木耳[Auricularia auricla(L.ex Hook)Underw]菌株为试材,对黑木耳总DNA提取方法和ISSR-PCR扩增反应条件及引物进行了筛选.结果表明.改进的SDS抽提法可以在较短时间内提取到高纯度的DNA产物;并筛选出了可产生遗传多样性的15条引物,其中引物ISSR11、ISSR12、ISSR14扩增结果的稳定性好、多态性强.经凝胶电泳比较,引物ISSR14可作为黑木耳PCR-ISSR反应的适宜引物.

The methods of DNA extration,primers of ISSR and rection conditions were selected using fourteen kinds of A uricularia auricular ag experiment materials.The results showed that the improved SDS method could obtain pure DNA in short time.Fifteen primers of genetic variation were selected.Primers ISSR11、ISSR12、ISSR14 could produce stable and intense polymorphism.Primer ISSR14 could be used as the suitable primer for A.auricula variety.
