目的 通过三代繁殖试验,观察外源性核苷酸对于啮齿类大鼠生育能力和后代生理、神经指标发育的影响,从而做出发育安全性评价. 方法 SD大鼠(FD代)断乳后,喂核苷酸,含量分别为0、0.01%、0.04%、0.16%、0.64%、1.28%的受试物饲料90 d后雌、雄1:1交配,所产仔鼠为F1;F1断乳后喂养90 d后进行二代繁殖试验.重复上述实验方法 直到多代繁殖实验结束,F3代出生;期间观察各亲代大鼠孕期、分娩、哺乳期情况和各子代哺乳期情况、生理发育指标、神经反射指标. 结果 外源补充核苷酸并未使亲代大鼠的孕期、分娩、哺乳期以及子代大鼠的哺乳期、生理和神经发育反射出现异常情况. 结论 在本实验剂量下,补充外源补充核苷酸对于多代大鼠的生殖功能和胚胎期、胎儿期以及哺乳期的发育是安全的.

Objective To investigate the effect of exogenous nucleotides on fertility in parents rats and the indexes of physiological and neural development in newborn rots over the three - reproductive exper-iment, and evaluate the safety on development. Methods Weanling rats (F0) were fed to exogenous nucleotides at the dose of 0, 0. 01%, 0. 04 %, 0.16 %,0.64 %, 1.28 % for 90 days and then male and female rats mated for 1 : 1 pattern. The offspring of F0 was F1. F1 rats were fed to exogenous nucleotides for 90 days after they were weaned. And then these rants go on to the second reproductive experiment. Repeat the progress above until F3 rats were born. In the whole period, we observed the gestational, partal, lacta-tional period in parent rats and the lactatioual period and the indexes of physiological and neural development in newborn rats. Results No abnormal situation was found among the gestatioual, partal, lactatioual pe-riod in parent rats and the lactational period and the indexes of physiological and neural development in new-born rats. Conclusion At the dose of the experiment, supplementing exogenous nucleotides was safe to reproductive function and embryonic, fetal, lactational period in multi -generation rats.
