目的 研究DNA跨损伤修复基因Rev1、Pol ζ(Rev3和Rev7)、Pol η、Pol ι、和Pol κ在脑胶质瘤中的表达,探讨这些基因的表达水平与脑胶质瘤发生、发展的关系.方法 利用SYBR Green实时定量PCR技术检测DNA跨损伤修复基因在85例原发脑胶质瘤病人和14例正常脑组织中的mRNA表达情况.结果 与正常脑组织相比,Rev1基因在Ⅳ级胶质瘤中表达上调(ΔCt为5.55±1.12,P<0.05);Rev3基因在Ⅱ级、Ⅲ级、Ⅳ级胶质瘤中表达上调(ΔCt分别为5.46±1.35、5.13±1.53和6.14±1.10,均P<0.05);Rev7基因在Ⅱ级、Ⅲ级、Ⅳ级胶质瘤中表达上调(ΔCt分别为5.88±0.82、5.41±1.43和5.56±0.96,均P<0.05);Pol η基因在Ⅱ级、Ⅲ级、Ⅳ级胶质瘤中表达上调(ΔCt分别为5.97±1.15、5.83±1.03和6.34±1.01,均P<0.05);Pol ι基因在Ⅱ~Ⅳ级胶质瘤中表达上调(ΔCt分别为6.67±1.17、6.33±1.39和7.03±1.06,均P<0.05);Pol κ基因在Ⅳ级胶质瘤中表达上调(ΔCt为5.25±1.58,P<0.05).Spearman秩相关分析表明,跨损伤修复基因Rev1、Rev3、Pol η、Pol ι和Pol κ的表达量与肿瘤病理学分级呈正相关(均P<0.01).结论 DNA跨损伤修复基因可能与胶质瘤的发生、进展相关.

Objective To detect the expressions of Rev1, Pol ζ, Pol η, Pol ι and Pol κ mRNA in human primary glioma tissues, and to analyze the relationship between the expression and the pathological grade.Methods We applied SYBR Green real-time PCR to investigate the mRNA expressions of the 6 genes in 85 primary glioma tissues and 14 normal brain tissues.Results The expressions of 4 genes including Rev3, Rev7, Pol η and Pol ι were significantly(P<0.05)higher in glioma tissues in grade Ⅱ, Ⅲ and Ⅳ as compared to normal brain tissues. The expressions of Rev1 and Pol κ were significantly(P<0.05)higher in grade Ⅳ glioma compared to the normal brain tissue. The positive correlation existed (P<0.01) between the expression of all genes mRNA and the pathologic grade of the human glioma, except for Rev7.Conclusion The up-regulation of DNA translesion synthesis genes, including Rev1, Rev3, Rev7, Pol η, Pol ι and Pol κ, may be associated with pathogenesis of glioma.
