用荧光分光光度法研究了盐酸洛美沙星在血清中的荧光光谱特性,并对其在血清样品中的含量进行了测定.用乙腈沉淀蛋白对血清进行预处理,在硼酸盐缓冲溶液中,用硫酸调节酸度,并使其pH值为3.0,以β环糊精为荧光增敏剂.通过扫描其激发光谱和发射光谱,可知盐酸洛美沙星在血清中有以下荧光特性:盐酸洛美沙星血清提取液和盐酸洛美沙星的标准溶液具有相同特征的激发光谱和发射光谱,其激发波长为288nm,发射波长为450nm,也即药物光谱不受血清成分干扰,但血清样品对盐酸洛美沙星的荧光强度造成强烈的荧光猝灭.测定血清中盐酸洛美沙星的线性范围为:0.v5~5.0μg·ml,其回归方程为:y=-0.237 0+4.809X(R=0.996 8),血清中盐酸洛美沙星的平均回收率为:99.5%,方法的相对标准偏差为:0.82%(n=5).
用熒光分光光度法研究瞭鹽痠洛美沙星在血清中的熒光光譜特性,併對其在血清樣品中的含量進行瞭測定.用乙腈沉澱蛋白對血清進行預處理,在硼痠鹽緩遲溶液中,用硫痠調節痠度,併使其pH值為3.0,以β環糊精為熒光增敏劑.通過掃描其激髮光譜和髮射光譜,可知鹽痠洛美沙星在血清中有以下熒光特性:鹽痠洛美沙星血清提取液和鹽痠洛美沙星的標準溶液具有相同特徵的激髮光譜和髮射光譜,其激髮波長為288nm,髮射波長為450nm,也即藥物光譜不受血清成分榦擾,但血清樣品對鹽痠洛美沙星的熒光彊度造成彊烈的熒光猝滅.測定血清中鹽痠洛美沙星的線性範圍為:0.v5~5.0μg·ml,其迴歸方程為:y=-0.237 0+4.809X(R=0.996 8),血清中鹽痠洛美沙星的平均迴收率為:99.5%,方法的相對標準偏差為:0.82%(n=5).

A method of florescence spectrophotometry for study of fluorescence spectrum characteristic and determination of lomefloxacin hydrochloride is developed in serum by florescence spectrophotometry. Acetonitrile is used to remove protein in the serum. The pH of the system is controlled at 3.0 in borate and vitriol buffer and the β-CD is added to boost up fluorescence. Through scanning excitation and emission spectrum, the fluorescence spectrophotometry of lomefloxacin hydrochloride in serum is known as follow: extracting solution of lomefloxacin hydrochloride in serum and standard solution of lomefloxacin hydrochloride owns the same character of excitation and emission spectrum, λex,m=288nm, λem,m=450nm. That is the spectrum of the drug is not distrubed by the components of serum, but the serum can quench lomefloxacin hydrochloride. The linear range is 0. 005~5.0μg/ml and regression equation is Y=-0. 237 0+4. 809X(R=0. 996 8). The average recovery of lomefloxacin hydrochloride is 99. 5% in serum. The relative standard deviation of the method is 0.82%(n=5).
