利用农杆菌介导法,将半夏凝集素(pta)基因和高赖氨酸蛋白(sb401)基因导入水稻中,经潮霉素抗性筛选,得到70株独立再生植株.通过PCR检测,从中筛选出18株转基因植株.对这些转基因植株当代进行Southern blotting分析表明:pta 基因和sb401基因已经共转化并整和到受体基因组中.对转基因植株后代的叶片进行RT-PCR分析表明:pta基因在转录水平上得到了有效的表达.测定10株T0转基因植株成熟种子的赖氨酸和总蛋白质量分数表明,与对照相比,赖氨酸和总蛋白的提高率分别为8.48%~35.34%和4.55%~32.74%,表明sb401基因在大部分转基因植株成熟种子中的蛋白质水平上得到了比较有效的表达.

The pta and sb401 genes were simultaneously transferred to rice in this study. Eighteen positive transgenic plants were identified by PCR. Southern blot assays for transgenic plants indicated that the two target genes have successfully been introduced and co-integrated into the genome of the receptors. RT-PCR analyses demonstrated the pta gene has expressed at the transcription level. The test of lysine and total protein content in mature seeds showed that, compared to the non-transgenic rice control, the content of lysine and total protein has respectively increased by 8.48~34.35 and 4.55~32.74 percent. The results confirmed that the sb401 gene has expressed at the translation level in rice seeds.
