[目的]比较2种方法进行桫椤atpB-rbcL序列测定的测序效果.[方法]通过PCR产物直接测序和克隆测序对桫椤叶绿体atpB-rbcL基因间隔序列进行测定与分析.[结果]结果表明,桫椤atpB-rbcL序列直接测序所得峰形紊乱,PolyT后出现多重峰;克隆测序结果峰形清晰,无干扰.[结论]对于桫椤atpB-rbcL序列测定,克隆测序优于直接测序.

[Objective]The research aimed to study the sequencing effects of atpB-rbcL sequence of Alsophila spinulosa with two different methods.[Method]The results of direct sequencing of PCR products or sequencing by cloning PCR products of atpB-rbcL uncoding region in A.spinuosa were compared and analyzed.[Result]The results showed that the profile of direct sequencing was disordered and overlapped after PolyT.The profile of cloned sequencing was clear and did not overlap.[Conclusion]Direct sequencing of PCR products was better than sequencing by cloning PCR products of the atpB-rbcL sequence in A.spinulosa.
