目的 探讨导致恶性肿瘤出现细胞染色体非整倍数现象的着丝粒蛋白-H(CENP-H)在舌鳞癌中的表达情况,及其对舌鳞癌细胞增殖的影响.方法 舌鳞状细胞癌组织及其癌旁舌黏膜组织组成配对子,采用RT-PCR、Western blot分别检测8对配对子的舌鳞癌系TSCCa、Tca8113及原代培养的舌黏膜细胞中CENP-H mRNA水平和蛋白的表达水平;采用免疫组织化学SP法检测同样8对配对子舌黏膜中CENP-H的表达;采用MTT法、克隆形成实验法分析转染CENP-H对TSCCa细胞增殖能力的影响.结果 舌鳞状细胞癌中CENP-H在mRNA、蛋白质水平的表达高于舌黏膜上皮,且差异有统计学意义;免疫组化显示CENP-H高表达,且阳性染色定位于肿瘤细胞的细胞核,而在舌黏膜鳞状上皮细胞中未见表达;MTT法、克隆形成试验显示转染CENP-H后舌鳞癌细胞增殖能力明显高于未转染的对照组细胞(P < 0.001).结论 舌鳞癌细胞中高表达的CENP-H对舌鳞癌细胞增殖起到促进作用,在舌鳞癌的发生发展中可能扮演重要角色.
目的 探討導緻噁性腫瘤齣現細胞染色體非整倍數現象的著絲粒蛋白-H(CENP-H)在舌鱗癌中的錶達情況,及其對舌鱗癌細胞增殖的影響.方法 舌鱗狀細胞癌組織及其癌徬舌黏膜組織組成配對子,採用RT-PCR、Western blot分彆檢測8對配對子的舌鱗癌繫TSCCa、Tca8113及原代培養的舌黏膜細胞中CENP-H mRNA水平和蛋白的錶達水平;採用免疫組織化學SP法檢測同樣8對配對子舌黏膜中CENP-H的錶達;採用MTT法、剋隆形成實驗法分析轉染CENP-H對TSCCa細胞增殖能力的影響.結果 舌鱗狀細胞癌中CENP-H在mRNA、蛋白質水平的錶達高于舌黏膜上皮,且差異有統計學意義;免疫組化顯示CENP-H高錶達,且暘性染色定位于腫瘤細胞的細胞覈,而在舌黏膜鱗狀上皮細胞中未見錶達;MTT法、剋隆形成試驗顯示轉染CENP-H後舌鱗癌細胞增殖能力明顯高于未轉染的對照組細胞(P < 0.001).結論 舌鱗癌細胞中高錶達的CENP-H對舌鱗癌細胞增殖起到促進作用,在舌鱗癌的髮生髮展中可能扮縯重要角色.

Objective To investigate the expression pattern and promoting proliferation function of centromere protein H (CENP-H) in Squamous cell carcinoma of the tongue which has been demonstrated to be associated with aneuploidy in carcinoma cell. Methods Reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot analysis were performed to test the expression of CENP-H in eight matched pairs of Squamous cell carcinoma of the tongue tissues (C) and adjacent noncancerous tongue tissues (N), as well as Squamous cell carcinoma of the tongue(TSCC) cell lines and tongue epithelial cells (TEC), the level of CENP-H in 8 characterized TSCC tissues was determined by immunohistochemistry. MTT assay, colony formation assay displayed that TSCCa with high expression of CENP-H reproductive activity. Results The mRNA and protein level of CENP-H was much higher in cancer tissues (C) and TSCC cell lines than that in adjacent noncancerous tongue tissues (N) and in normal cells, respectively. MTT assay, colony formation assay displayed that high expression of CENP-H promoted the proliferation of TSCC cell line TSCCa (P < 0.001). Conclusion CENP-H is highly expressed in TSCC, and its high expression promotes proliferation.
