为了提高冬小麦花培苗染色体加倍效率,分别用不同浓度的秋水仙碱对参试的冬小麦材料的花药愈伤组织、再生植株根系和花培苗分蘖节进行了加倍处理。结果表明,用0.02‰和0.05‰秋水仙碱浓度处理的愈伤组织再生植株结实率达33.3%~61.5%。用0.2%的秋水仙碱浸根处理5h,结实株率平均高达37.5%。用0.04%的秋水仙碱1%的二甲亚砜溶液浸泡分蘖节的时间应在5~10h之间较为适宜,结实株率平均可达50%以上。
為瞭提高鼕小麥花培苗染色體加倍效率,分彆用不同濃度的鞦水仙堿對參試的鼕小麥材料的花藥愈傷組織、再生植株根繫和花培苗分蘗節進行瞭加倍處理。結果錶明,用0.02‰和0.05‰鞦水仙堿濃度處理的愈傷組織再生植株結實率達33.3%~61.5%。用0.2%的鞦水仙堿浸根處理5h,結實株率平均高達37.5%。用0.04%的鞦水仙堿1%的二甲亞砜溶液浸泡分蘗節的時間應在5~10h之間較為適宜,結實株率平均可達50%以上。
위료제고동소맥화배묘염색체가배효솔,분별용불동농도적추수선감대삼시적동소맥재료적화약유상조직、재생식주근계화화배묘분얼절진행료가배처리。결과표명,용0.02‰화0.05‰추수선감농도처리적유상조직재생식주결실솔체33.3%~61.5%。용0.2%적추수선감침근처리5h,결실주솔평균고체37.5%。용0.04%적추수선감1%적이갑아풍용액침포분얼절적시간응재5~10h지간교위괄의,결실주솔평균가체50%이상。
In order to increase chromosome doubling frequency for anther culture of winter wheat, the callus, roots of regenerative plants and tillering node were treated respectively with different concentration of colchicine. The result showed that there were 33.3%~61.5% setting percentage in the treated callus with 0.02‰ and 0.05‰ colchicine soluion. After soaking roots of regenerative plants for 5 hours using 0.02‰ and 0.05‰ colchicine soluion, the average setting percentage reached 37.5%. The setting percentage were over 50% in tillering node soaked for 5~10 hours using 0.04‰ colchicine soluion and 1‰dimethylsulfoxide.
