目的 探讨骨髓造血细胞(HC)移植在大鼠实验性结肠炎(EC)模型的炎性反应肠管和正常脏器的定植情况.方法 健康雌性SD大鼠54只,完全随机分为移植组(A组)、模型对照组(B组)和非模型对照组(C组),每组均为18只.A组和B组采用三硝基苯磺酸(TNBS)法建立EC模型,C组用生理盐水灌肠.用悬浮培养法在体外培养获得雄性SD大鼠的HC,移植前24 h用5-溴-2-脱氧尿嘧啶(BrdU)标记HC.A组造模后24 h将含HC的细胞悬液经尾静脉注入大鼠体内;B、C组则同法注射生理盐水作对照.每组于移植后第7天、第14天及第2l天处死大鼠各6只,取全段结肠及肝脏、胃、胰腺组织送检,用免疫组化方法检测HC的定植情况.结果 BrdU阳性细胞在A组结肠组织各时间段均有定植(100%),移植后第14天和第2l天的病变区阳性细胞比例显著高于同时间段非病变区(P<0.05).A组的18例肝组织中有3例亦可检测到BrdU阳性细胞(16.7%).而胃、胰腺组织未检测到阳性细胞,两对照组均阴性.结论 同种异体HC移植能在EC大鼠模型的肠道及肝脏中定植,HC的定植与EC病变的严重程度和局部血供丰富情况均有关系.

Objective To investigate the localization of allogeneic bone marrow hematopoietic cells (HC) transplantation in experimental colitis (EC) and normal organs in rat models.Methods Fifty-four healthy female SD rats were complete randomly divided into three groups: the transplantation group (group A, n=18), model control group (group B, n=18) and non-model control group (group C, n=18).HC from allogeneic SD rats were culture- expanded in vitro and labeled with BrdU 24 hours before transplantation, and transplanted into the TNBS-induced rat models of EC by tail vein injection (group A).Saline was administrated in EC rata (group B) and normal rats received saline enemas (group C) by tail vein injection.Six rats were put to death in each group at days 7, 14, and 21 after transplantation.The BrdU immunoreactive cells were detected in the whole colon and liver, stomach and pancreas tissues of three groups by immunohistochemical staining.Results In group A, the BrdU immunoreactive cells were present in the colons at day 7, 14, and 21.Ratio of pesitive cells was higher in lesions than that in normal tissues at day 14, and 21 (P<0.05).Most of them were located in the mucous epithelium layers.In group A, immunoreactive cells could also be found in liver tissues (3/18, 16.7%), but were not found in stomach and pancrease tissues.No BrdU immunoreactive cells were found in colons of the controls (groups B and C).Conclusion Allogeneic HC transplantation can be localized in colons and livers of TNBS-induced rat models of EC.The localization of HC is related to the degree of EC and local blood supply.
