目的:观察丹参素对血小板衍生生长因子-BB(Platelet derived growth factor-BB,PDGF-BB)刺激大鼠肝星状细胞(Hepatic stellate cell,HSC)增殖、Ⅰ型胶原合成、磷酸化细胞外信号调节激酶1,2(Phosphoryhted extracellular signal-regulated kinase 1/2,P-ERK1/2)表达的影响,探讨丹参素抗肝纤维化的分子机制.方法:四甲基偶氮唑盐法检测大鼠HSC增殖活性;免疫细胞化学染色法测定大鼠HSC Ⅰ型胶原合成的表达;Western blot测定大鼠肝星状细胞P-ERK1/2的表达.结果:丹参素有抑制PDGF-BB刺激大鼠HSC增殖,Ⅰ型胶原表达,p-ERK1/2表达的作用.结论:丹参素可抑制PDGF-BB刺激的大鼠HSC增殖,Ⅰ型胶原合成,其机制可能与抑制ERK1/2信号转导通路有关.

목적:관찰단삼소대혈소판연생생장인자-BB(Platelet derived growth factor-BB,PDGF-BB)자격대서간성상세포(Hepatic stellate cell,HSC)증식、Ⅰ형효원합성、린산화세포외신호조절격매1,2(Phosphoryhted extracellular signal-regulated kinase 1/2,P-ERK1/2)표체적영향,탐토단삼소항간섬유화적분자궤제.방법:사갑기우담서염법검측대서HSC증식활성;면역세포화학염색법측정대서HSC Ⅰ형효원합성적표체;Western blot측정대서간성상세포P-ERK1/2적표체.결과:단삼소유억제PDGF-BB자격대서HSC증식,Ⅰ형효원표체,p-ERK1/2표체적작용.결론:단삼소가억제PDGF-BB자격적대서HSC증식,Ⅰ형효원합성,기궤제가능여억제ERK1/2신호전도통로유관.
Objective:To investigate the inhibitory effect of Danshengsu on proliferation, collagen type Ⅰ synthesis and Phosphorylated extracellular signal- regulated kinase 1/2 (P-ERK1/2)expression in rat hepatic stellate cell (HSC) stimulated by platelet derived growth factor-BB (PDGF-BB), and to elucidate the molecular mechanism of Danshensu against hepatic fibrosis: Methods: Cell proliferation was evaluated by 3-(4,5-dimethylthiazol-2-yl )-2,5-Diphennyltetrazolium bromide(MTT)colorimetric assay.Collagen type Ⅰ synthesis was detected by Immunocytochemical stain.Phosphorylated extracellular signal- regulated kinase 1/2 (P-ERK1/2) expression was detected by Western blot. Results:Danshensu could inhibit proliferation,collagen type Ⅰ synthesis and P-ERK1/2 expression of HSC stimulated by PDGF-BB. Conclusion:Danshensu can inhibit proliferation and Collagen type Ⅰ synthesis of HSC stimulated by PDGF-BB,the mechanism of which may be Danshensu, s inhibitory effect on ERK1/2 signal transduction pathway.
