目的 观察基质金属蛋白酶-9(MMP-9)和色素上皮衍生因子(PEDF)基因表达在胰腺癌(PC)侵袭转移中的作用。方法 应用免疫组织化学法检测35例胰腺癌标本中癌细胞的MMP-9和PEDF的表达,统计分析两种分子标记物与胰腺癌细胞MMP-9和PEDF的表达。结果 胰腺癌组中MMP-9和PEDF表达率分别为71.4%和34.3%,MMP-9高表达率和PEDF低表达率与浸润范围、淋巴转移和远处转移呈正相关(P<0.05)。MMP-9与PEDF表达呈负相关(P<0.01)。结论 MMP-9在胰腺癌侵袭转移中起促进作用,PEDF可抑制肿瘤生长,PEDF低表达可能通过上调MMP-9的表达,从而促进胰腺癌的侵袭转移。

목적 관찰기질금속단백매-9(MMP-9)화색소상피연생인자(PEDF)기인표체재이선암(PC)침습전이중적작용。방법 응용면역조직화학법검측35례이선암표본중암세포적MMP-9화PEDF적표체,통계분석량충분자표기물여이선암세포MMP-9화PEDF적표체。결과 이선암조중MMP-9화PEDF표체솔분별위71.4%화34.3%,MMP-9고표체솔화PEDF저표체솔여침윤범위、림파전이화원처전이정정상관(P<0.05)。MMP-9여PEDF표체정부상관(P<0.01)。결론 MMP-9재이선암침습전이중기촉진작용,PEDF가억제종류생장,PEDF저표체가능통과상조MMP-9적표체,종이촉진이선암적침습전이。
Objective To investigate the expression of matrix metalloproteinase-9 (MMP-9) and pigment epithelium-derived factor (PEDF) and their correlation with tumor invasion and metastasis in human pancreatic carcinoma. Methods Immunohistochemistry was used to detect the expression of MMP-9 and PEDF in 35 cases of pancreatic carcinoma tissue samples. The relationship between the molecular markers and tumor invasion and metastasis was analyzed. ResultsThe expression rate of MMP-9 and PEDF was 71.4% and 34. 3% respectively in pancreatic carcinoma patients. MMP-9 expression was related to depth of invasion and lymph node metastasis as well as distant metastasis. Low PEDF protein expression was significantly associated with tumor-increased penetration, lymph node and distant metastasis.There was a negative correlation between the expression of MMP-9 and PEDF. Conclusion MMP-9 can accelerate the development and progression of pancreatic carcinoma, while PEDF play a negative role in tumor genesis and progress. The low expression of PEDF may promote the tumor invasion by upregulating the expression of MMP-9.
