本研究以l0-4M碘化钠(NaI)刺激单层培养的人Graves病(GD)甲状腺滤泡上皮细胞(TEC),采用定性及半定量PCR技术检测刺激前后白介素-6(IL-6)、γ-干扰素(IFN-γ)和肿瘤坏死因子-a(TNF-a)在细胞中的表达水平。结果表明:(1)3例GD组织均含有IL-6及IFN-γ mRNA,2例检测到TNF-a基因表达;(2)基础状态下,3例TEC均表达IL-6基因,2例表达TNF-a,而所有样本均无IFN-γ mRNA;(3)Nal不能诱导TEC产生IFN-γ mRNA,对IL-6 mRNA的表达亦无明显影响;(4)1例TNF-a阴性的TEC样本,经NaI刺激后,可表达mRNA,2例原含有TNF-a的TEC经刺激后,其mRNA的表达水平显著增加。提示碘可通过诱导或增强TEC表达TNF-a,导致自身免疫性甲状腺疾病的发生与发展。

본연구이l0-4M전화납(NaI)자격단층배양적인Graves병(GD)갑상선려포상피세포(TEC),채용정성급반정량PCR기술검측자격전후백개소-6(IL-6)、γ-간우소(IFN-γ)화종류배사인자-a(TNF-a)재세포중적표체수평。결과표명:(1)3례GD조직균함유IL-6급IFN-γ mRNA,2례검측도TNF-a기인표체;(2)기출상태하,3례TEC균표체IL-6기인,2례표체TNF-a,이소유양본균무IFN-γ mRNA;(3)Nal불능유도TEC산생IFN-γ mRNA,대IL-6 mRNA적표체역무명현영향;(4)1례TNF-a음성적TEC양본,경NaI자격후,가표체mRNA,2례원함유TNF-a적TEC경자격후,기mRNA적표체수평현저증가。제시전가통과유도혹증강TEC표체TNF-a,도치자신면역성갑상선질병적발생여발전。
To investigate the influence of iodide on cytokine gene expression in thyroid cells and the effect of iodide in the pathoggenesis of autoimmune thyroid diseases (AITD). Thymocytes were cultured in the absence or presence of 10-4MNaI. Interleukin-6 (IL-6), tumor necrosis factor-a (TNF-a)and interferon-γ (IFN-γ)mRNA levels in thyroid cell were detected by semiquantitative reverse transcription polymerase chain reaction (RT-PCR). Results showed: (1)IL-6,TNF-a and IFN-γ were detected in 3/3, 2/3 and 0/3 unstimulated cell samples, respectively. (2)NaI did not induce gene expression of IFN-γ in thyroid epithelial cells (TEC), and could not stimulate IL-6 mRNA expression , while TNF-a mRNA level was significantliy increased by NaI. It concludes that iodide exerts its important roles in the initiation and progression of AITD through TNF-a production.
