[目的] 快速测定大花蕙兰和墨兰花粉活力,保证人工杂交育种有效开展. [方法] 以大花蕙兰和墨兰花粉为材料,用离体培养法和TTC染色法分别测定花粉活力,同时进行不同温度贮藏试验.[结果] TTC染色法所测得的花粉活力均略高于离体培养法,TTC对花粉染色12 h即可达到较好的观察效果; -20和-80 ℃冷藏花粉1年后仍保持较高活力.[结论] TTC法操作简便,需时短,可作为大花蕙兰和墨兰花粉活力测定便捷、有效的方法.-20 ℃低温、干燥条件可作为大花蕙兰和墨兰花粉保存的简易方法.

[Objective] The research aimed to test the pollen viability to improve the probability of crossing.[Method] The pollen of Cymbidium hybridum and Cymbidium sinense cultivars were used as materials,and two methods were compared in pollen viability testing between in vitro germination and TTC dyeing.The pollen was stored at different conditions.[Result] Pollen viability by TTC was higher than that by in vitro germination,but there was no significance between them.The pollen could be at high viability after storing at the environment of -20 ℃ or -80 ℃ for more than one year.[Conclusion] TTC dyeing could be used as an effective method to test the orchid pollen viability because of saving time and convenience.Storing at –20 ℃ while dry could be chosen as a feasible approach for orchid pollen when preparation for cross breeding.
