目的:利用微生物转化的方法对大黄中的游离蒽醌类化合物进行结构修饰.方法:筛选了21种微生物对大黄酚(1),大黄素甲醚(2),大黄素(3)的转化作用.结果表明刺囊毛酶对大黄酚,大黄素甲醚,大黄素具有转化作用.结果:制备、分离刺囊毛酶对大黄酚,大黄素甲醚,大黄素的转化产物,分别得到大黄酚-8-O-β-D-葡萄糖苷(4),大黄素甲醚-8-O-β-D-葡萄糖苷(5),大黄素甲醚-1-O-β-D-葡萄糖苷(6),ω-羟基大黄素(7)4个转化产物.利用半乳糖代替葡萄糖作为培养基的碳源,制备、分离刺囊毛酶对大黄酚的转化产物,初步考察了培养基中不同的碳源对糖苷化产物的影响.得到的转化产物为大黄酚-1-O-β-D-葡萄糖苷和大黄酚-8-O-β-D-葡萄糖苷(8a和8b)的混合物,而非大黄酚的半乳糖苷.

AIM:To study the biotransformation for the structure modification of anthraquinones.METHOD:Mucor spinosus was employed for the biotransformation of three rhubarb anthraquinones-chrysophanol(1),physcion(2) and emodin(3).RESULT AND CONCLUSION:Four metabolites were obtained. On the basis of their physio-chemical data,the structures of the biotransformated products were characterized as chrysophanol-8-O-β-D- glucopyranoside(4),physcion-8-O-β-D-glucopyranoside(5),physcion-1-O-β-D- glucopyranoside(6) and citreorosein(7) respectively. Compound 1 was incubated in the culture of Mucor spinosus by using galactose as carbon source instead of glucose,it was found that the corresponding product was a mixture of chrysophanol-1-O-β-D-glucopyranoside and chrysophanol-8-O-β-D-glucopyranoside(8a and 8b) other than the corresponding galactose glycoside as expected. By this test,we preliminarily explore with the influence of different kinds of carbon sources added to the medium on the glycosylation.
