Lycium barbarum in northeast%polysaecharide%Isolation%purification%identification
研究了东北枸杞多糖的提取、分离纯化及其组分和结构的鉴定,结果表明:枸杞多糖水提的最佳条件为提取温度100℃,pH值11,料液比1:20,浸提2h;通过DEAE-SephadexA-50柱层析得到纯度较高的LBP-4,薄层分析LBP-4主要是由鼠李糖、木糖、葡萄糖、甘露糖和半乳糖醛酸组成.红外光谱和刚果红实验分析LBP-4具有糖类的特征吸收峰,是多种单糖通过α糖苷键及β糖苷键连接组成的杂多糖,不具有三股螺旋结构.

Isolation, purification and identification of Lycium barbarum polysaccharide in northeasarea were studied in this paper.The result showed that the optimum extraction technology of LBP was the extraction temperature 100℃,pH=11, ratio of material to water 1:20, the extraction time 2h. DEAE-SephadexA-50 purification could obtain high purity LBP-4. The analysis of TLC indicated that LBP-4 was mainly made up of Xyl., Man., Glu. and galactose acid. IR spectrum and Congo red analysis showed that LBP-4 possessed the characteristics of carbohydrate absorption peak, which is a eomplex polysaccharides composed of sugars linking through ot-glucoside lindage and β glucoside without three spiral structure.
