目的 探讨急性吗啡暴露对大鼠前额叶皮质τ蛋白磷酸化及学习记忆的影响.方法 40只雄性SD大鼠随机均分成急性吗啡暴露组(M组)和对照组(C组).M组单次腹腔注射吗啡(30 mg/kg),C组同法注射等量生理盐水.注射后2 h和24 h分别断头处死10只大鼠.免疫印迹技术检测前额叶皮质r蛋白的磷酸化水平.后处死的大鼠于注射后2、4及24 h分别行水迷宫实验.结果 M组注射后2 h时平台寻找时间显著长于C组(P<0.01).M组大鼠前额叶皮质τ蛋白Ser396和Thr231位点的磷酸化水平于注射后2 h时显著高于C组(P<0.05).结论 急性吗啡暴露可致大鼠学习记忆功能可逆性减退,前额叶皮质r蛋白过度磷酸化可能与其有关.
目的 探討急性嗎啡暴露對大鼠前額葉皮質τ蛋白燐痠化及學習記憶的影響.方法 40隻雄性SD大鼠隨機均分成急性嗎啡暴露組(M組)和對照組(C組).M組單次腹腔註射嗎啡(30 mg/kg),C組同法註射等量生理鹽水.註射後2 h和24 h分彆斷頭處死10隻大鼠.免疫印跡技術檢測前額葉皮質r蛋白的燐痠化水平.後處死的大鼠于註射後2、4及24 h分彆行水迷宮實驗.結果 M組註射後2 h時平檯尋找時間顯著長于C組(P<0.01).M組大鼠前額葉皮質τ蛋白Ser396和Thr231位點的燐痠化水平于註射後2 h時顯著高于C組(P<0.05).結論 急性嗎啡暴露可緻大鼠學習記憶功能可逆性減退,前額葉皮質r蛋白過度燐痠化可能與其有關.

Objective To explore the effect of acute morphine on memory and phosphorylation of the prefrontal cortex of rats. Methods Forty SD rats were randomly divided into two groups with 20 rats each. Group M received intraperitoneally injection of morphine 30 mg/kg. Group C was given normal saline instead of morphine as the control. Ten rats from each group were sacrificed to get the prefrontal cortex for detecting τ protein phosphorylation at the sites Ser396 and Thr231 with immunoblotting at 2 h and 24 h after injections. Laterly sacrificed rats in each group were tested with morris water maze(MWM) at 2 h,4 h and 24 h after injections. Results The latent period was significantly longer in group M than that of group C(P<0. 01) at 2 h after injections. The hyperphosphorylation at the sites of Ser396 and Thr231 of τ in group M was significantly higher than that in group C (P< 0. 05) at 2 h after injection. Conclusion The intraperitoneally injection of morphine 30 mg/ kg may lead to transient memory decrease in rats, which may be related to the hyperphosphorylation at the sites of Ser396 and Thr231of τ in the prefrontal cortex.
