目的:观察2型糖尿病患者脂肪细胞内胰岛素信号转导蛋白:胰岛素受体底物-1(Insulin receptor substrate-1,IRS-1)酪氨酸磷酸化及蛋白激酶B(Protein kinase B,PKB/AKT)ser473磷酸化水平以及微炎症因子白细胞介素6(Interleukin-6,IL-6)的表达.方法:取2型糖尿病患者(2型糖尿病组)及非糖尿病患者(非糖尿病对照组)腹部皮下脂肪组织原代培养,用免疫沉淀法及蛋白印迹法(Western blot)检测两组脂肪细胞内胰岛素刺激后IRS-1酪氨酸磷酸化及AKT-Ser473磷酸化程度,酶联免疫吸附法检测脂肪细胞培养液中IL-6蛋白的水平.结果:(1)2型糖尿病组脂肪细胞内胰岛素刺激后IRS-1酪氨酸磷酸化及AKT-Ser473磷酸化水平明显低于非糖尿病对照组,差异有统计学意义(P<0.05);(2)2型糖尿病组脂肪细胞培养液中IL-6蛋白的水平明显高于非糖尿病对照组,差异有统计学意义(P<0.05).结论:2型糖尿病患者脂肪细胞存在胰岛素抵抗和微炎症状态.

목적:관찰2형당뇨병환자지방세포내이도소신호전도단백:이도소수체저물-1(Insulin receptor substrate-1,IRS-1)락안산린산화급단백격매B(Protein kinase B,PKB/AKT)ser473린산화수평이급미염증인자백세포개소6(Interleukin-6,IL-6)적표체.방법:취2형당뇨병환자(2형당뇨병조)급비당뇨병환자(비당뇨병대조조)복부피하지방조직원대배양,용면역침정법급단백인적법(Western blot)검측량조지방세포내이도소자격후IRS-1락안산린산화급AKT-Ser473린산화정도,매련면역흡부법검측지방세포배양액중IL-6단백적수평.결과:(1)2형당뇨병조지방세포내이도소자격후IRS-1락안산린산화급AKT-Ser473린산화수평명현저우비당뇨병대조조,차이유통계학의의(P<0.05);(2)2형당뇨병조지방세포배양액중IL-6단백적수평명현고우비당뇨병대조조,차이유통계학의의(P<0.05).결론:2형당뇨병환자지방세포존재이도소저항화미염증상태.
Objective: To investigate the insulin signal transduction protein in the adipocytes of Type 2 diabetics: Insulin Receptor Substrate-1(IRS-1) tyrosine phosphorylation and Protein Kinase B (PKB/AKT) Sei473 phosphorylation level and expression of microinilammatory oytokine interleukin-6( IL-6 ). Methods: Using primary cell culture, adipocytes from abdominal subcutaneous adipose tissue of Type 2 diabetics(Type 2 diabetes group) and non-diabetics (non-diabetes control group) were cultured.IRS-1 tyrosine phosphorylation and AKT-Ser473 phosphorylation level in the adipocytes of the two groups with insulin stimulation was detected by the Immunoprecipitation and western blot IL-6 level in adipocytes culture supernatant was evaluated by ELISA. Results: (1) The IRS-1 tyrosine phosphorylation and AKT-Ser473 phosphorylation level in the adipocytes with insulin stimulation of Type 2 diabetes group were significantly lower than that of the non-diabetes control group (P<0.05); (2) The IL-6 protein level in the adipocytes medium of Type 2 diabetes group was significantly higher than that of the non-diabetes control group (P< 0.05 ). Conclusion: The adipocytes of Type 2 diabetics present insulin resistance and microinflammatory states.
