目的 研究大鼠脑源性骨质疏松的特性.方法 用雌性SD大鼠建立脑源性骨质疏松动物模型,用体内双荧光标记法测其右胫骨上段骨组织形态计量学动态参数,用XLA27153型流式细胞仪测定CD_3、CD_4、CD_8和NK细胞的百分率(%).用~3H-TdR掺入法测定大鼠体外T淋巴细胞转化功能.用放射免疫法测定血清中生长素(GH)、雌二醇(E_2)和骨钙素(BGP)的含量.用酶法测定血脂,称体脂质量.结果 与对照组比较,模型组大鼠:(1)代表骨形成的骨组织形态计量学动态参数值显著降低:荧光标记周长百分率(%L.PM),骨矿化沉积率(MAR),骨形成率(BFR/BS,BFR/BV)分别降低了33.6%(P<0.01),25.5%(P<0.01),48.0%(P<0.01)和46.3%(P<0.01);(2)生长素(GH)、雌二醇(E_2)和骨钙素(BGP)的含量显著降低;(3)免疫功能下降:CD_3、CD_4、CD_8、CD_4/CD_8、NK细胞百分率明显降低和体外T淋巴细胞转化能力明显降低;(4)脂质代谢紊乱:TC,TG,LDL-C显著升高,HDL-C显著降低,体脂质量显著增加.结论 大鼠脑源性骨质疏松是一种多因素的综合的神经、内分泌、免疫功能和脂质代谢功能减退和紊乱的低转换型骨质疏松.

Objective To investigate the characteristics of the osteoporosis of brain origin in rats.Methods (1) The model of osteoporosis of Brain Origin in female rats was established.(2) Morphological methods were used to estimate the bone histomorphometry parameters (3)Radioimmunoassay (RIA) was used to measure the level of the serum growth hormone (GH),estradiol (E_2) and bone Ga protein (BGP).(4) CD_3 (%),CD_4 (%),CD_8(%),CD_4/CD_8 (%) and NKC (%) were measured in rat peripheral blood.(5) ~3H-TdR was used to measure the changes of convertibility of T lymphocytes in rat peripheral blood.(6) Serum lipids and body fat weight were measured.Results Compared with control group,the model group rats:(1) Bone formation parameters decreased:% L.PM,MAR,BFR/BS,BFR/BV were decreased by33.6%(P<0.01),25.5%(P<0.01),48.0%(P<0.01) and 46.3% (P < 0.01) respectively;(2) The levels of serum E_2,GH,BGP significantly decreased;(3) CD_3,(%),CD_4 (%),CD_8 (%),CD_4/CD_8 (%) and NKC (%) significantly decreased,and convertibility of T lymphocytes decreased obviously;(4) Levels of total cholesterol (TC),triglyceride (TG),and low density lipoprotein-cholesterol (LDL-C) significantly increased,but concentration of high density lipoprotein-cholesterol (HDL-C) decreased obviously,and body fat weight significantly increased.Conclusion The osteoporosis of brain origin is the one of low bone turnover osteoporosis with the decrease of GH,E_2,BGP and immune function,and lipid metabolism disorder as well.
